摘 要:目的 :探讨一氧化氮 (NO)在肝移植急性反应中的功能作用及 NO合酶 (NOS)的细胞定位。方法 :应用金黄地鼠至大鼠异种原位肝移植急性排斥反应及大鼠同基因原位肝移植动物模型 ,观察 L - NMMA对受体存活期、AL T、TNF-α及移植肝的病理影响 ,应用 NADPH - d组化染色观察 NOS的活性及其细胞来源。结果 :表明 L - NMMA可明显降低异种肝移植组受体存活期、加剧其肝功能恶化、上调 TNF-α、加剧移植肝的病理损害 ;异种肝移植组 NADPH- d组化染色呈强阳性 ,主要在肝实质细胞及浸润炎性细胞表达。结论Objective:To investigate the role of nitric oxide in acute rejection of golden hamster to rat orthotopic liver xenotransplantation and the cellular origin of nitric oxide synthase. Methods:By establishing the model of golden hamster to rat orthotopic liver xenotransplantation following acute rejection and Wistar rat to rat syngeneic orthotopic liver transplantation,we observed the effect of NO synthase inhibitor(L NMMA) on the recipient survival,ALT,TNF α and pathology of grafted liver. Using NADPH diaphorase histochemistry staining,we studied the activities of NO synthase and cellular origin of nitric oxide. Results:Administration of NO synthase inhibitor(L NMMA) can markedly reduce the recipient survival,deteriorate the grafted liver function,upregulate the expression of TNF α,gravitated the pathological injury of the grafted liver. The expression of nitric oxide synthase was primarily present in both hepatocytes and portal inflammatory infiltrate cells for NADPH diaphorase histochemistry staining in orthotopic liver xenotransplantation.Conclusion:These results suggest that nitric oxide may play a potential immunologic protective role within the settings of acute rejection of golden hamster to rat orthtopic liver xenotransplantation.
