摘 要:100只24周龄健康海兰褐蛋鸡随机分为2组,分别喂以对照日粮(日粮中含钙3.55%)和低钙日粮(日粮中含钙1.27%)60d。用改良150g/L EDTA脱钙液制备骨组织切片,以正常蛋鸡胫骨为参照,观察笼养蛋鸡骨质疏松症病鸡胫骨病理组织学和显微结构的变化。结果表明,该改良骨组织脱钙法缩短了脱钙时间,骨组织切片薄而平整,骨组织结构清晰,HE染色对比鲜明;与对照组相比,低钙组蛋鸡胫骨中的皮质骨出现许多吸收腔,在吸收腔内有大量破骨细胞,髓质骨小梁周围破骨细胞增多,骨吸收作用明显增强,导致骨质疏松。表明本方法在笼养蛋鸡骨质疏松症的病理诊断方面具有很好的应用价值。100 24-week-old hyline brown laying hens were randomly divided into two groups and ted on di- ets as control diet (containing 3.55% calcium) and low calcium diet (containing 1.27% calcium) respec- tively for 60 days. Tibiae were decalcified in the modified decalcifying fluid (150 g/L EDTA), and then de- calcified tibia sections were made for observing the changes of histopathology and microstructure in tibia of laying hens with osteoporosis compared with the controls. The results showed that decalcification duration was short, the section samples were thin, even and integrated, and the bone structure was clear with H E staining by the modified method for making bone sections. Compared with control group, many resorp- tion cavities within the cortical bone were present together with an increase in number of osteoclasts in these cavities and around the medullary bone trabeculae in low calcium group, osteoclastic bone resorption was markedly increased and led to osteoporosis. The study showed that the modified decalcification of bone tissue had good application value in the pathologic diagnosis of cage layer osteoporosis.
