梅山猪SLC36A1基因cDNA克隆、组织表达及生物信息学分析

摘　　要：为了研究SLC36A1基因的功能及其与梅山猪毛色等表现的关系,试验以梅山猪皮肤组织cDNA为模板,PCR扩增SLC36A1基因完整CDS序列,利用生物信息学方法分析SLC36A1基因CDS序列及其编码的氨基酸序列,用RT-PCR方法检测SLC36A1基因在梅山猪各组织中的相对表达量。结果表明:SLC36A1基因CDS全长为1 431 bp,编码476个氨基酸,属于跨膜疏水性蛋白,具有转运功能,是比较保守的基因;RT-PCR检测该基因在梅山猪雄性生殖系统及肾脏中有较高的表达量,在肝脏、肌肉、子宫、输卵管等组织中表达量很低,并且首次发现该基因在尿道球腺中有最高的表达量。To study the function of the SLC36A1 gene and its relationship with the manifestation of coat color, the complete coding sequence （CDS） of the SLC36A1 gene was obtained by PCR amplification using the eDNA from the skin tissue of Meishan pig as a template. The CDS of the SLC36A1 gene and its encoded amino acid sequence were analyzed by bioinformatics method, and then the RT - PCR assay was used to determine the relative expression level of the SLC36A1 gene in various tissues of Meishan pig. The results showed the CDS of the SLC36A1 gene was 1 431 bp in size, encoding for a protein of 476 amino acids. The encoded protein was a transmembrance hydrophobic protein with the func- tion of transport function. The SLC36A1 gene belonged to a relatively conserved gene. It was found that the SLC36AI genes had higher expres- sion levels in kidney and male reproductive system of Meishan pig, but had lower expression levels in liver, muscle, uterus, oviduct, and so on. It was first found that the SLC36AI gene had the highest expression level in the bulbourethral gland.
