摘  要:目的采用经典的二维电泳方法建立不同耐药表型鲍氏不动杆菌的蛋白质差异表达图谱,为后续从整体水平探讨耐药机制奠定试验基础。方法对来自医院临床分离并鉴定的不同耐药表型鲍氏不动杆菌采用VITEK-2Compact进行药敏检测,同时用纸片扩散法(K-B法)检测头孢哌酮/舒巴坦、阿米卡星、米诺环素的敏感性;按药敏试验表型分为:全耐药菌株、全敏感菌株、头孢哌酮/舒巴坦及米诺环素敏感菌株;然后进行二维电泳,之后用Bradford法进行蛋白点的显色。结果与全敏感鲍氏不动杆菌相比,其余耐药表型有3个共同蛋白点表达明显减低或是缺失,同时有1个共同蛋白点表达明显增加;头孢哌酮/舒巴坦敏感菌株与全耐药菌株和米诺环素敏感菌株相比,在某个区域存在多个差异蛋白点;米诺环素敏感菌株与全耐药和头孢哌酮/舒巴坦敏感菌株相比,在另外区域也存在多个差异蛋白点。结论初步建立了不同耐药表型的鲍氏不动杆菌双向电泳图谱,为后续的差异蛋白的鉴定和功能研究提供了试验依据。OBJECTIVE To establish distinguished protein expression profiles of different drug resistance pheno- types of Acinetobacter baumannii by using the classic two-dimensional electrophoresis method so as to provide an experimental basis for further study of drug resistance mechanism from the whole level. METHODS The drug sus- ceptibility testing for the clinical isolates of different drug resistance phenotypes of A. baumannii was performed by VITEK-Compact, then the drug susceptibility of the strains to cefoperazone-sulbactam, amikaein, or minocycline was tested by means of the K-B method~ the strains, according to the result of drug susceptibility testing, were di- vided into the following groups: the fully drug-resistant strains, fully drug-susceptible strains, cefoperazone-sul- bactam-susceptible strains, minocycline-susceptible strains; the two-dimensional electrophoresis assay was per- formed, then the ehromogenic protein spots were observed with the use of Bradford method. RESULTS Compared with the fully drug-susceptible strains, three common protein spots expressions of other drug resistance pheno- types of strains were reduced or disappeared obviously, and one common protein spot expression was increased sig- nificantly; compared with fully drug-resistant strains and the minocycline-susceptible strains, the eefoperazone-sul- bactam-susceptible strains showed multiple diverse protein spots in a certain region; compared with the fully drug- resistant strains and the cefoperazone-sulbactam-suseeptible strains, only minocycline-susceptible strains displayed multiple diverse protein spots in other regions. CONCLUSION The two-dimensional electrophoresis profiles for the different drug resistance phenotypes of A. baumannii haven been preliminarily established, which may provide ex- perimental basis for the subsequent identification of protein and the function study.
