摘  要:目的复制稳定的大鼠糖尿病膀胱病模型。方法将SD大鼠随机分为实验组和对照组,实验组大鼠随机分为A、B、C组,分别一次性腹腔注射50、60和70 mg/kg的链脲佐菌素(STZ),对照组大鼠注射等量柠檬酸钠缓冲液,每隔1周观察各组大鼠的体质量、空腹尾静脉血糖变化、成模及存活情况,连续续观察8周,第8周检测各组的尿流动力学,尿流动力学检测后取膀胱组织做石蜡病理切片,HE染色,光镜下观察结果。结果一次性腹腔注射STZ 60 mg/kg组的成模率最高(80%),存活率高(86.7%),死亡率较低(13.3%),其逼尿肌收缩功能较对照组弱:最大膀胱容量(4.14±0.46)mL,而对照组为(2.15±0.42)mL(P<0.05);最大膀胱压(2.81±0.61)kPa,而对照组为(5.10±0.72)kPa(P<0.05);残余尿(2.14±0.46)mL,而对照组为(0.22±0.09)mL(P<0.05);膀胱顺应性(1.61±0.08)mL/kPa,而对照组为(0.73±0.06)mL/kPa(P<0.05)。结论一次性腹腔注射STZ60mg/kg是复制稳定大鼠糖尿病膀胱病模型的最佳剂量。[Objective] To establish stable diabetic neurogenic bladder model in rat. [Methods] SD rats were randomly divided into experimental and control groups, the experimental rats were randomly divided into three groups for single intraperitoneal injection of 50 mg/kg, 60 mg/kg, 70 mg/kg Streptozotocin (STZ) respec tively, the control group was treated with the equal amount of sodium citrate buffer. Each group was observed for their mortality, body weight, the successful molded rate and instantaneous blood glucose test in tail vein at every other week after injection, and eight weeks of continuous observation. Followed urodynamic testing on 8 week, the bladders were separated for paraffin section and HE staining. [Results] The single 60 mg/kg STZ injection group showed the highest survival rates (80%) and successful molded rate (86.7%), lower mortality rates (13.3%), and weakened detrusor contraction function compared with the control group: the maximum bladder capacity (P〈0.05), compliance (P〈0.05) and residual urine (P〈0.05) in models were found signifi cantly higher than control. However, the maximum bladder pressure significantly decreased comparing with that of the control group (P〈0.05). [ Conclusion] A single intraperitoneal injection of 60 mg/kg STZ is the ideal dose for inducing the stable rat model of diabetic bladder disease.
