膳食锌含量对小鼠注射乙肝疫苗后免疫应答的影响

摘  要:目的评估膳食中锌含量与小鼠注射乙肝疫苗后免疫应答之间的关系。方法 45只BALB/c小鼠随机分为3组,分别接受指定的不同锌含量饮食。经过4周喂养后,每只BALB/c小鼠胫前肌内分别注射100μL重组酵母菌乙肝疫苗,2周及4周后分别加强注射1次,加强注射后2周处死小鼠,检测小鼠淋巴细胞刺激指数及血清中乙型肝炎病毒表面抗体(HBsAb)水平,比较各组之间差异。结果 10.0μg/mL乙型肝炎病毒表面抗原(HBsAg)诱导后各组的刺激指数ZD组(2.88±0.18),ZA组(4.86±0.53),ZE组(3.99±0.46)。ZA组与ZD组比较差异有统计学意义(t=3.007,P=0.006),而与ZE组比较差异没有统计学意义(t=1.912,P=0.07)。ZD组HBsAb水平在3组中最低。ZA、ZE组中位抗体水平较ZD组升高,具有统计学意义(P<0.01),有更多的小鼠抗体水平达到或超过10 000 IU/L。ZA、ZE组间比较无明显差异(P>0.05)。结论锌缺乏膳食小鼠注射乙肝疫苗后,无论是体外细胞介导的免疫应答还是体内特异性的体液免疫应答均下降。膳食锌含量可影响小鼠接种乙肝疫苗后机体的免疫应答效力。[Objective] To evaluate the relationship between Zinc content in diet and post-vaccination immune response to hepatitis B in mice. [Methods] Forty-five BALB/c mice, were randomly dirided into three groups and received the assigned diets throughout the entire study period. After 4 weeks of feeding, BALB/c mice were immunized intramuscularly at the tibialis anterior muscles with 100 μL/mouse yeast-de rived hepatitis B surface antigen (HBsAg), followed 2 weeks and 4 weeks after the first injection by a single booster dose. Serum samples were collected by retro-orhital bleeding after 2 weeks following the second im munization. The stimulation index and hepatitis B surface antibody (HBsAb) were measured, the differences a mong each groups were analyzed in order to illuminate the relationship between Zinc content in diet and post-vaccination immune responsiveness to hepatitis B in mice. [Results] The stimulation index incubated in the presence of 10.0 μg/mL HBsAg was determined in all groups of mice as follows: ZD (2.88 ± 0.18), ZA (4.86 ± 0.53), ZE (3.99 ± 0.46). Compared with ZA and ZE, stimulation index of ZD decreased significantly (t = 3.007, P=0.O06). There were not significantly different between ZA and ZE (t=1.912, P=0.07). Mice receiving the Zinc deficient diet demonstrated the lowest titer of all groups tested. ZA and ZE were significantly higher than ZD (P〈0.01). Mice receiving Zinc adequacy or zinc excess diets demonstrated an increase in the median antibody titer, with more mice achieving titers at or greater than 10,000 IU/L. No statistically significant differ- ence could be seen among ZA and ZE (P〉0.05). [Conclusion] Both cell-mediated immune responses in vitro and hepatitis B surface antigen specific antibody responses in vivo following hepatitis B vaccine are decreasedin mice fed a Zinc deficient diet. Zinc content in diet could effect post-vaccination immune responsiveness to hepatitis B in mice.
