摘  要:根据刺五加法呢基焦磷酸合酶(farnesyl diphosphate synthase,FPS)基因的cDNA序列信息,设计特异性引物,利用半定量RT-PCR技术,对刺五加FPS基因在不同产地、不同器官、不同发育时期及茉莉酸甲酯(MeJA)处理后的表达情况进行分析。结果表明,FPS基因在不同产地、时期、器官及MeJA处理后的刺五加中均有表达,但表达量存在显著差异。FPS在萌芽期的表达量最高,盛花期的表达量最低,两者比值为4.19且差异显著;本溪、珲春、鸡西、雾灵山、穆棱、梅河口、伊通产地刺五加的FPS相对表达量依次降低;在不同器官中以叶的表达量最高;MeJA处理后FPS的表达量比用蒸馏水处理的对照组均有显著提高。FPS的相对表达量与刺五加叶器官的皂苷含量存在显著的正相关关系。Primers were designed according to the available sequence information of the cloned FPS(farnesyl diphosphate synthase) gene of Eleutherococcus senticosus. Semi-quantitative RT- PCR was developed to analyze relative expression quantity of FPS gene during different growth, different dates, different organs,and the organs treated with MeJA. The results showed that FPS gene was expressed but with significant difference at expression levels. FPS had a highest expres- sion level during the germination stage and the lowest level during the full flowering stage. The ratio of the highest level to the lowest level was 4.19,with a significant difference. FPS relative expression reduced in order of the individuals from Benxi, Hunchun,Jixi, Wulingshan Mountain, Muling, Meihekou, and Yitong. The highest expression value of FPS was in leaf blade. After trea- ted with MeJA,FPS relative expression increased and was higher than that of the control group. There was significantly positive correlation between the content of Eleutherococcus senticosus sap- onins and the expression levels of FPS gene.
