摘  要:目的观察GLPP对Alzheimer样大鼠记忆能力和海马超微结构的影响。方法雄性Wistar大鼠60只,随机分为4组,每组15只:正常对照组、模型组、NS组、GLPP组(250 mg/kg);除正常对照组(正常昼夜节律)外,其余各组连续光照(光照度400Lux,24 h),其间GLPP组ig GLPP,每天1次;NS组ig相同体积的生理盐水,每天1次;光照30d后Morris水迷宫法测试各组大鼠空间记忆能力,透射电镜观察海马线粒体和突触结构的改变。结果模型组大鼠寻台潜伏期明显延长,与正常对照组比较有显著性差异(P<0.05);GLPP组寻台潜伏期明显缩短,与模型组比较有显著差异(P<0.05);透射电镜结果显示:正常对照组、GLPP组大鼠海马线粒体神经轴突和神经突触基本正常;模型组、NS组大鼠海马线粒体肿胀、结构紊乱,膜结构破坏,嵴模糊、消失;神经髓鞘内神经细丝稀少,神经突触缺失、减少,突触密度降低,突触间隙不清,突触小泡减少。结论 GLPP可增强Alzheimer样大鼠空间记忆能力,减轻持续光照对大鼠海马超微结构的破坏。Objective To observe the effect of ganoderma lucidum polysaccharides peptide (GLPP) on spatial memory and the mitochondrion, neural synapse in hippocampus of Alzheimer-like rats. Method 60 male Wistar rats were divided randomly into four groups (15 per group): normal controls, model group, NS group and GLPP group (250 mg/kg). Except for normal controls (received 12 : 12 hr light- dark cycle) , all other groups were received continuous illumination (400 lux illuminance) for 30 days. During the illuminating period, GLPP group was received GLPP (ig, one time per day) ; NS group was received normal saline of the same volume as the GLPP group ( ig, one time per day). After an illuminat- ing period of 30 days, Morris water maze test was used to measure the spatial memory of rats. Transmis- sion electron microscope (TEM) observed the changes of mitochondria and neural synapsis in hippocam- pus. Result Compared with normal controls, the spatial memory of model group was significantly re- duced (P 〈 0.05). Compared with model controls, the spatial memory of GLPP group was significantly enhanced (P 〈 0.05). The results by the transmission electron microscope showed that normal control group and GLPP group beared normal mitochondria and synapse. However, for the model group and NS group, mitochondria were swelling, structure in hippocampus was destroyed, mitochondrial crista was disappearance. Decrease of neurofilaments within the nerve myelin, the density of neural synapsis was decreased and synaptic vesicle was decresed. Conclusion GLPP can reduce the spatial memory and protect mitochondria and synapses in the hippocampus from the damage caused by continuous illumination and defect of Alzheimer-like rat.
