摘  要:目的研究锰对大鼠体内生殖激素和乳酸脱氢酶(LDH)水平的影响,探讨锰对大鼠生精细胞caspase-3表达与生殖激素和LDH水平之间的关系。方法 48只雄性SD大鼠,体质量(120±10)g随机分为6组:空白对照组,低剂量(15 mg/kg MnCl2)和高剂量(30 mg/kg MnCl2)组,8只/组。MnCl2组分别染锰4 w和6 w,空白对照组给予等量NS,给药途径均为腹腔注射,5 d/w,1次/d。大鼠分别于第4w末和第6w末处死。免疫组化法检测睾丸组织中caspase-3表达,放射免疫法测定血清T、FSH和LH含量,化学比色法测定血清和睾丸LDH活性。结果与空白对照组比较,各染锰组caspase-3阳性细胞率均显著升高,血清T含量显著降低,FSH和LH含量显著升高,6 w 30mg/kg组血清LDH活性显著升高,各染锰组睾丸LDH活性显著降低(P<0.01)。染锰剂量相同,6 w与4 w比较,caspase-3阳性细胞率和血清FSH、LH含量显著升高(P<0.01),血清和睾丸LDH活性分别显著升高和降低(P<0.01)。染锰时间相同,高剂量组与低剂量组比较,caspase-3阳性细胞率和血清FSH、LH含量均显著升高(P<0.01),血清和睾丸LDH活性分别显著升高和降低(P<0.01)。结论染锰15 mg/kg 4 w即可诱发大鼠血清T含量降低,FSH和LH含量升高及睾丸LDH活性降低和血清LDH活性升高,可能是促进生精细胞caspase-3表达的重要原因。Objective The purpose was to study the expression of caspase-3 in spermatogenic cells and the contents of reproductive hormone, lactate dehydrogenase (LDH) of Rats Exposed to Manganese and to discuss the relationship between them. Method 48 male rats weighting (120 ± 10) g were randomly divided into 6 groups (blank control, 15 mg/kg and 30 mg/kg MnCl2) , 8 rats/group. MnCl2 groups were contaminated manganese (MnCl2 · 4H2O) by I. P. for 4 and 6 weeks respectively and blank control groups was injected with NS. All rats were injected once a day, five times per week. In the 4th and 6th weekend, their testis and serum were collected. The expression of caspase-3 in testis was investi- gated by immunohisto chemical methods, and the contents of T, FSH and LH in serum were measured by radioimmu noassay, the activity of LDH in serum and testis were measured by colorimetric analysis. Results Compared with blank control groups, the caspase-3-positive-cell rate was increased,the contents of T was decreased but FSH and LH were increased in every manganese exposure group (P 〈 0.01 ) , the activity of LDH was increased in serum in 6 w 15 mg/kg group, and decreased in testis in every MnCl2 groups(P 〈 0.01 ). Among manganese exposure groups of same dose, the caspase-3-positive-cell rate and the contents of FSH, LH in serum were increased, the activity of LDH was increased in serum but decreased in testis significantly in 6 w MnCI~ groups than it in 4 w groups (P 〈 0.01 ). Among manganese exposure groups of same time, the caspase-3-positive-cell rate and the contents of FSH,LH in serum were increased, the activity of LDH was increased in serum but decreased in testis in 30 mg/kg manganese exposure groups than them in 15 mg/kg groups (P 〈 0.01 ). Conclusion MnCl2 at dose of 15mg/kg can decrease the contents of T, increase the contents of FSH, LH in serum, and decrease the activity of LDH in testis and increase it in serum, and they might be the important causes of accelerating the expression
