摘 要:过渡金属配合物催化DNA的断裂反应是近年来最为活跃的前沿研究领域之一,研究发现某些金属配合物具有催化DNA裂解的功能,因而该研究对新型抗肿瘤药物的设计及其生物学的研究具有重要意义和应用前景.为了研究金属与DNA的作用机制,在室温条件下,采用溶剂挥发法合成配合物[Ni(BDA)(H2O)4]2H2O(其中BDA为2,2'-联吡啶-4,4'二羧酸),并应用红外光谱、元素分析和X-射线衍射技术对配合物的结构进行表征,所有表征均证明合成的是目标配合物.通过凝胶电泳法测定配合物对pBR322质粒DNA的切割能力,发现配合物对pBR322质粒DNA具有一定的切割能力.利用MTT法检测配合物对HeLa细胞的抑制活性,结果表明该配合物对HeLa细胞具有一定的抑制能力.Chain cleavage reaction of DNA catalyzed by transition metal complexes is one of the most active forefront research fields in recent years. Studies have found that some metal complexes have special functions in catalyzing DNA cleavage, so this research area is significant and has application prospect on the design of new anticancer drugs and biology activity research. In order to investigate the mechanism of metal and DNA, we synthesized the complex [ Ni (BDA) ( H2 O) 4 ] 2H2O ( BDA for 2,2'-bipyridyl-4,4'- dicarboxylic acid) by the solvent evaporation method at room temperature, and applied infrared spectra, elemental analysis and X-ray diffraction technology to characterize the structure of the complex. Gel elec- trophoresis assay demonstrate the ability of the complex to cleave the pBR322 plasmid DNA. The MTT method was used to detect the complexes for inhibitory activity of HeLa cell, and results showed that the complex had inhibition on HeLa cell.
