摘 要:目的研究促红细胞生成素衍生肽(HBSP)对顺铂所致大鼠急性肾损伤(AKI)的肾脏保护作用的剂量反应关系,进一步探讨HBSP在AKI的使用方法。方法清洁级健康SD大鼠随机分为5组,分别为:对照组(5只)、顺铂组(20只)、HBSP(8 nmol/kg)组(20只)、HBSP(16 nmol/kg)组(20只)、HBSP(32 nmol/kg)组(20只)。应用全自动生化分析仪检测大鼠血尿素氮、肌酐、血红蛋白,苏木素-伊红(HE)染色观察肾组织的病理变化及TUNEL法检测肾小管上皮细胞凋亡。结果各组大鼠不同时间(实验前及实验的第1、3、5、7天)血尿素氮及肌酐水平比较,差异均有统计学意义(尿素氮:F组间=441.456,F时间=10.643,P<0.001;肌酐:F组间=417.933,F时间=9.412,P<0.000 1);各组大鼠不同时间血红蛋白水平比较,差异无统计学意义(F组间=566 645.213,F时间=1.581,P=0.200)。各组大鼠不同时间肾组织TUNEL染色百分比比较,差异有统计学意义(F组间=226.100,F时间=9.553,P<0.000 1)。结论 HBSP对顺铂所致AKI的肾脏保护作用可能与使用剂量有关。Objective To investigate dose response relationship between helix B surface peptide (HBSP, a kind of erythropoietin -derived peptide) and cisplatin -induced acute kidney injury (AKI) in rats, so as to the usage of HBSP in treat- ment of AKI could be further discussed. Methods Healthy Sprague - Dawly (SD) rats of clean grade were randomly divided into5 groups: control group (5), cisplatin group (20), HBSP group (8 nmol/kg) (20), HBSP group (16 nmol/kg) (:20), HBSP group (32 nmol/kg) (20) . Serum creatinine, urea nitrogen and hemoglobin were detected by automatic bio- chemistry analyzer, the renal tissues were stained by HE to reflect the degree of renal pathology, apoptosis of tubular epithelium was detected by TUNEL assay. Results At various time points ( before experiment and at 1, 3, 5, 7 day after experiment), there were significant differences in serum creatinine and urea nitrogen levels between groups (BUN: Fgroup = 441. 456, Ftime = 10. 643, P 〈0. 001 ; Scr: Fgroup =417. 933, Ftime = 9. 412, P 〈 0. 000 1 ), there was no significant difference in hemoglobin levels between groups (Fgroup = 566 645. 213, Ftime = 1.581, P = 0. 200), there was significant difference in percentages of kid- ney tissue TUNEL staining levels ( Fgroup = 226. 100, Ftime = 9. 553, P 〈 0. 000 1 ) . Conclusion The renal protective effects of HBSP on cisplatin - induced acute kidney injury may be related with HBSP doses.
