摘  要:以产酶活性与产酶稳定性为指标评价3种固定化细胞产壳聚糖酶(Chitosanase,EC3.2.1.99)方法,以酶比活力、酶结合效率、酶活力回收率及稳定性评价4种固定化壳聚糖酶方法,以筛选最优固定化方案应用于壳聚糖酶的生产。结果表明,海藻酸钠壳聚糖包埋法固定化细胞的产壳聚糖酶活性较高且稳定性好;海藻酸钠壳聚糖吸附包埋固定化壳聚糖酶的酶比活力、酶结合效率与酶活力回收率均为最高,且稳定性强。Chitosanase production was optimized through evaluating three methods of cells immobilization using the activity and stability of enzyme production as indicators. Four methods of enzyme immobilization were evaluated using of the enzyme combination efficiency, enzyme activity recovery and enzyme stability as indicators. The results showed that the sodium algi- nate and chitosan embed method was the optimal method of cells and enzyme immobilization due to its best performance in the activity and stability of enzyme production and in the enzyme activity, enzyme combination efficiency, enzyme activity re- covery and enzyme stability.
