摘 要:以近缘昆虫Hsp60基因保守区域设计兼并引物,PCR扩增烟粉虱(Bemisia tabaci)地中海(MED)隐种与温室白粉虱(Trialeurodes vaporariorum)Hsp60基因cDNA,并检测了Hsp60基因受温度影响的表达情况。结果表明,烟粉虱MED隐种Hsp60基因cDNA的开放性阅读框长1 821 bp,编码607个氨基酸;温室白粉虱Hsp60基因cDNA的开放性阅读框长1 788 bp,编码596个氨基酸,Hsp60基因在昆虫纲低级阶元水平和高级阶元水平系统进化上能得到一个较理想结果。温室白粉虱Hsp60基因表达量受温度影响显著(P<0.05),而烟粉虱MED隐种则不显著(P>0.05)。Primers were designed based on the reported sequences of conservative regions of Hsp60 gene family in other in- sects, and the fragments isolated from the two whitefly were amplified by PCR and the full-length cDNA sequences were obtained by sequence assembly. The full-length cDNA of Hsp60 of the two whitefly contained 1 821 and 1 788 bp, open reading frame(ORF) encoded 607 and 596 amino acids, respectively. Meanwhile, the results showed that the Hsp60 presented good phylo- genetic informativeness at both low and high taxonomic levels in insect. Trialeurodes vaporariorum Hsp60 gene expression was found significantly affected by temperature(P0.05), but Bemisia tabaci MED was not signficant(P0.05).
