摘  要:C4型PEPC基因导入C3植物可提高C3植物的光合能力,抑制油菜C3型PEPC基因的表达能提高油菜含油量,因此要进一步提高油菜的光合生产力和促进油脂积累,有必要在油菜叶片中特异性地表达玉米的C4型PEPC基因。通过克隆玉米、水稻和油菜的rbcS叶片特异性启动子,构建rbcS调控的玉米C4型PEPC基因叶片特异性表达载体,转化油菜获得叶片特异性高效表达的转PEPC基因植株,与原种和转玉米C4型PEPC基因油菜相比较,研究各株系PEPC酶活性和光合生理表现;根、茎、叶与种子器官玉米C4型PEPC基因的表达差异;分析各株系油菜油脂和蛋白质含量、单株产量等农艺性状。通过在油菜叶片中特异性地表达玉米的C4型PEPC基因,提高油菜单叶的光合速率,促进油菜油脂积累,为油菜的高产育种提供新途径。The C3 plant photosynthetic capacity was improved by introducing the C4-type PEPC gene. To modify the expression of PEPC gene in rape by co-suppression may facilitate the flow of phosphoenolpyruvate into lipid at times of high lipid accumulation.In order to further improve the photosynthetic capacity and oil contents in rape, leaf-specific expression of the C4-type PEPC gene in transgenic rape was necessary. rbcS promoter of maize, rice and rape was cloned and the vector of leaf-specific expression of the plant recombinants was constructed. The transgenic rape with leaf-specific expression of the C4-type PEPC gene was acquired by hypocotyl disseminated infection and floral-dip method. The PEPC activity, photosynthetic performance, differential expression of PEPC gene in root, stem, leaf and seed and protein and lipid contents in rape in transgenic rape would be studied. The photosynthetic rate and oil contents was improved by leaf-specific expression of the C4-type PEPC gene in transgenic rape, providing a new way for the high-yield breeding of rape.
