摘 要:探讨了组织块贴壁法及酶消化法(胰蛋白酶、胶原酶、胰蛋白酶-EDTA、胰蛋白酶-EDTA-胶原酶)对分离黄鳝(Monopterus albus)肝细胞的细胞数量、细胞活力和细胞增殖能力的影响。结果表明,酶消化法优于组织块贴壁法,其中胰蛋白酶-EDTA-胶原酶分次消化法所获得细胞数量最多,为(1.30±0.06)×107个/g,胰蛋白酶消化法所得细胞活力最高,为(90.2±1.2)%,胰蛋白酶-EDTA-胶原酶分次消化法所获得活细胞数量最多,为(1.10±0.05)×107个/g。细胞增殖能力呈先升后降的趋势,在48 h细胞数量最多,144 h后细胞数量明显下降。The influences of different methods including tissue block adhering wall method and enzyme digestions(trypsin, collagenase, trypsin-EDTA, trypsin-EDTA-collagenase) on the cell number, the cell vitality and the cell proliferation ability of hepatocytes from Monopterus albus were studied. The results showed that the enzyme digestions were better than the tissue block adhering wall method. The most number of cells were(1.30±0.06)×107piece / g acquired by trypsin-EDTA-collagenase di- gestion. The highest cell activity was(90.2±1.2)% obtained by trypsin digestion. The most number of living cells were(1.10± 0.05)×107piece / g acquired by trypsin-EDTA-collagenase digestion. Cell proliferation ability peaked at 48 h and decreased at 144 h.
