摘  要:为了检测肉质相关基因TCAP(Titin-cap,Telethonin)启动子与转录因子MyoD(Myogenic differentiation antigen)的体内结合情况,采用染色质免疫共沉淀(Chromatin immunoprecipitation,ChIP)结合PCR技术分析TCAP启动子与转录因子MyoD的结合。结果表明,以MyoD抗体免疫沉淀的DNA片段为模板,PCR扩增获得了TCAP基因启动区121 bp的片段,实现了转录因子MyoD与TCAP启动子DNA序列结合。试验证实TCAP基因是MyoD调控的下游基因,在肌肉发育过程中发挥重要作用。The aim of this research was to identify whether MyoD transcription factor binds with TCAP(Titin-cap, Telethonin) gene promoter. The interaction between MyoD transcription factor and TCAP gene promoter was detected by Chromatin im- munoprecipitation(ChIP) and PCR technologies. The results showed that the specific 121 bp fragment of TCAP gene were de- tected in the DNA fragments immunoprecipitated by antibody specific for MyoD. This indicated that MyoD transcription factor could bind with TCAP gene promoter. The TCAP gene is one of downstream genes regulated by MyoD, thus plays an impor- tant role in the process of muscle development.
