摘  要:目的探讨黄芪、丹参有效单体黄芪甲苷和丹参多酚酸A干预慢性肾功能衰竭(CRF)的可能作用机制。方法 40只SD大鼠通过5/6肾切除建立CRF动物模型,根据血肌酐(Cr)值随机分为模型组、黄芪甲苷组、丹参多酚酸A组及科素亚组各10只,并设正常组10只。黄芪甲苷组给予黄芪甲苷1.4mg/(kg·d),丹参多酚酸A组给予丹参多酚酸A 17.1mg/(kg·d),科素亚组给予科素亚8.6mg/(kg·d)灌胃,模型组和正常组予等量蒸馏水灌胃。8周后测定各组大鼠血清Cr、尿素氮(BUN)水平,并检测肾组织转化生长因子β1(TGF-β1)mRNA及结缔组织生长因子(CTGF)mRNA的表达。结果黄芪甲苷组、丹参多酚酸A组血清Cr、BUN较模型组显著降低(P<0.01)。黄芪甲苷组、丹参多酚酸A组、科素亚组肾组织TGF-β1、CTGF mRNA较模型组亦降低(P<0.05或P<0.01)。丹参多酚酸A组在降低BUN、Cr方面优于科素亚组(P<0.05或P<0.01)。结论黄芪甲苷、丹参多酚酸A可能通过抑制肾组织TGF-β1、CTGF mRNA的表达,降低血清Cr、BUN水平,从而延缓CRF肾功能衰竭的恶化。Objective To research the possible mechanism of Astragaloside and Salvianolateon A for chronic re- nal failure (CRF). Methods The CRF models were established by 5/6 nephrectomy in Sprague rats. The rat models were randomized into the model group, Astragaloside group, Salvianolateon A gro Dawley up and (SD) Cozaar group according to the serum creatinine (SCr) value, with 10 in each. Another 10 rats were selected as the normal group. The Astragaloside group, Salvianolateon A group and Cozaar group was gavaged with 1.4 mg/( kg · d) Astra- galoside, 17.1 mg/(kg· d) Salvianolateon A and 8.6mg/(kg · d) Cozaar respectively. The model group and nor- mal group were gavaged with the same amount of distilled water. The levels of SCr and blood urea nitrogen (BUN) were detected 8 weeks later. The expressions of transforming growth factor-β1 ( TGF-β1 ) and connective tissue growth factor (CTGF) mRNA were detected. Results Comparing with the model group, the levels of SCr and BUN in the Astragaloside group and Salvianolateon A group were significantly decreased ( P 〈 0. 01 ). Comparing with the model group, the expressions of TGF-I31 and CTGF mRNA in the Astragaloside group, Salvianolateon A group and Cozaar group were significantly decreased (P 〈 O. 05 or P 〈 O. O1 ). The Salvianolateon A group was superior to the Cozaar group in decreasing BUN and SCr (P 〈0. 05 or P 〈0. 01 ). Conclusion Astragaloside and Salvianolateon A can slow the progression of renal failure probably by inhibiting the expressions of TGF-β1 and CTGF mRNA and decrea- sing the levels of SCr and BUN.
