摘 要:目的观察五紫三黄方对醋酸泼尼松片干预治疗的哮喘模型小鼠下丘脑—垂体—肾上腺(HPA)轴功能的影响。方法将120只BALB/C小鼠随机分为空白组、模型组、醋酸泼尼松片组和五紫三黄方低、中、高剂量组,每组20只。空白组小鼠用生理盐水腹腔注射和雾化,其余5组小鼠于第1、7、14天给予腹腔注射10μg卵蛋白+2mg氢氧化铝凝胶;第21天开始用1%卵蛋白雾化激发,每天1次,连续7天,其后每隔3天雾化1次,共雾化18次。每次雾化前醋酸泼尼松片组和五紫三黄方低、中、高剂量组小鼠均给予3.9 mg/kg醋酸泼尼松灌胃,第42天开始按照每两天醋酸泼尼松0.325 mg/kg速度撤减激素用量,于第63天全部撤离;同时从第35天开始五紫三黄方低、中、高剂量组分别给予五紫三黄方配方颗粒1.994g/kg、3.988 g/kg、7.976 g/kg灌胃,模型组和空白组给予等体积的生理盐水;第64天采用放射免疫法测定各组小鼠血浆促肾上腺皮质素释放激素(CRH)、促肾上腺皮质激素(ACTH)和皮质酮(COR)含量。结果模型组与空白组小鼠血浆COR、CRH和ACTH含量比较差异无统计学意义(P>0.05);与模型组比较,醋酸泼尼松片组小鼠以上指标均降低(P<0.05或P<0.01);与醋酸泼尼松片组比较,五紫三黄方低、中、高剂量组以上指标均有升高(P<0.05或P<0.01),且五紫三黄方高剂量组的改善程度优于五紫三黄方低、中剂量组(P<0.05或P<0.01)。结论五紫三黄方能够改善醋酸泼尼松片干预治疗哮喘小鼠导致的HPA轴功能受损,且大剂量使用效果更佳。Objective To observe the effect of Wuzi Sanhuang Formula [ formula of Ziheche (Placenta Hominis,, Zidanshen (Radix et Rhizoma Salviae Miltiorrhizae,, Ziyuan (Radix et Rhizoma Asteris,Zisuzi ( Fructus Perillae,Zishiying ( Fluoritum,Huangjing ( Rhizoma Polygonati,黄精)Huangqin (Radix Scutellariae, 黄芩) and Mahuang ( Herba Ephedrae, 麻黄) ] on the function of hypothalamic- pituitary-adrenal (HPA) axis in asthmatic mice intervened with prednisone. Methods Totally 120 BALB/C mice were randomized into the blank group, model group, Prednisone Acetate Tablets group and Wuzi Sanhuang Formula low-dose, middle-dose and high-dose groups, with 20 in each. The blank group was given intraperitoneal injection and atomization of normal saline. The other five groups were given intraperitoneal injection of 10 I^g ovalbumin and 2 mg aluminum hydroxide gel on the 1st, 7th and 14th day and 1% ovalbumin atomization from the 21st day once daily for 7 consecutive days and once every three days with a total of 18 times. The Prednisone Acetate Tablets group and Wuzi Sanhuang Formula low-dose, middle-dose and high-dose groups were garaged with 3.9 mg/kg prednisone acetate before each atomization. The steroid was withdrawn from the 42nd day at a dose of 0. 325 mg/kg every other day and completely withdrawn on the 63rd day. The Wuzi Sanhuang Formula low-dose, middle-dose and high-dose groups were garaged with 1. 994 g/kg, 3.988 g/kg and 7. 976 g/'kg l^uzi Sanhuang Formula granules and the model group and blank group were gavaged with the same amount of normal saline from the 35th day. The contents of corti- cotropin releasing hormone (CRH), adrenocorticotropic hormone (ACTH) and corticosterone (COR) were detected with radioimmunoassay on the 64th day. Results There was no signficant difference between the model group and blank group in the contents of COR, CRH and ACTH ( P 〉 0. 05 ).
