摘 要:目的建立同时测定咪达唑仑(MID)、硝西泮(NZP)、地西泮(DZP)3种药物血药浓度的反相高效液相色谱法。方法血浆样品以乙酸乙酯+石油醚提取,色谱柱为Symmetry shield RP18柱(250 mm×4.6 mm,5.0μm),流动相为甲醇-磷酸盐缓冲液(55∶45,v/v,pH2.15),流速为0.9 mL/min,检测波长为230 nm,柱温为26℃,进样量为20μL。结果 MID,NZP,DZP质量浓度均在20~4 000 ng/mL范围内与峰面积呈良好线性关系(r≥0.999 6,n=5);最低检测浓度(S/N=3),NZP,DZP为10 ng/mL,MID为20 ng/mL;回收率均在98.2%~103.9%之间,日内及日间RSD均<5%。结论该法能够同时测定3种药物的血药浓度,且简便、准确,适用于临床血药浓度监测。Objective To simultaneously determine the plasma concentrations of midazolam(MID), nitrazepam(NZP) and diazepam (DZP) by RP- HPLC. Methods The plasma sample was extracted with ethyl acetate and petroleum ether. The chromatographic column was the Symmetry shield RP^s column(250 mmx4.6 mm, 5.0 ~m) with the mobile phase of methanol-phosphate buffer(55:45, v/v, pH 2. 15) and the flow rate of 0. 9 mL/min; the detection wavelength was set at 230 nm; the column temperature was 26 ~C;the injection volume was 20 vLL. Results The mass concentration of MID, NZP and DZP within the range of 20 -4 000 ng/mL showed good linear relation with the peak area(r~〉0. 999 6, n =5). The minimal detectable concentration of NZP and DZP was l0 ng/mL, which of MID was 20 ng/mL (S~ N = 3). The recovery rate was 98.2% -103.9%. RSD of within- day and between- day was 〈 5%. Conclusion The method can simultaneously determine the plasma concentration the these 3 kinds of antiepileptic drug, which is convenient, accurate and suitable to monitor the plasma drug concentration.
