罗格列酮对阿霉素肾硬化大鼠整合素连接激酶及纤维连接蛋白表达的影响

摘　　要：目的研究罗格列酮对阿霉素肾硬化大鼠整合素连接酶和纤维连接蛋白表达的影响。方法选取60只3月龄体质量在200～250g雄性SD大鼠,随机分为3组,阿霉素硬化组20只、治疗组20只、对照组20只;硬化组大鼠行左肾切除术,术后注射阿霉素,治疗组在阿霉素硬化组基础上于肾切除当天行罗格列酮灌胃,对照组行假手术和尾静脉注射等量生理盐水;第10周末处死大鼠,观察各组大鼠肾脏光镜和电镜下的病理变化,计算肾小球系膜增生率和肾小球硬化率;肾组织标本采用免疫荧光方法检测各组整合素连接激酶(ILK)及纤维连接蛋白(Fn)的表达;RT-PCR及Western blot检测各组ILK及Fn的表达。结果成功建立肾硬化大鼠模型,硬化组肾硬化程度、ILK、Fn表达较对照组增加,治疗组肾硬化程度、ILK、Fn表达较肾硬化组减少,上述结果差异均有统计学意义(P<0.05)。结论整合素连接激酶和纤维连接蛋白水平的升高对肾硬化有一定的提示作用,罗格列酮对肾硬化的控制作用良好,可为临床治疗肾硬化提供一定的参考。OBJECTIVE To study the impact of rosiglitazone on expression of integrin-linked kinase（ILK）and fibronectin（Fn）in adriamycin rats.METHODS 60male SD rats with three months of age and body weight varying between 200and 250g were enrolled in the study and randomly divided into three groups：the adriamycin sclerosis group（20rats）,the treatment group（20rats）,the control group（20rats）,then the left kidney resection was performed in the adriamycin sclerosis group and doxorubicin was injected after the surgery;the rosiglitazone gavage was performed in the treatment group on the basis of the adriamycin sclerosis group on the day of nephrectomy,while the sham surgery was performed and normal saline was injected to the tail vein in the control group;the rats were killed at the tenth week,the renal pathological changes were observed under the light microscopy and electron microscopy,the glomerular mesangial proliferation rate and the rate of glomerular sclerosis were calculated;the kidney tissues were sampled to detect the expression of ILK and Fn by means of immunofluorescence assay;the RT-PCR and Western blot were performed to detect the expression of ILK and Fn.RESULTS The rat model of renal sclerosis has been successfully established;the renal sclerosis degree and the expression of ILK and Fn of the adriamycin sclerosis group were increased as compared with the control group;while the renal sclerosis degree and the expression of ILK and Fn were decreased as compared with the adriamycin sclerosis group,the differences were significant（P0.05）.CONCLUSIONThe elevated level of ILK and Fn plays an certain role of indicating the renal sclerosis,and rosiglitazone can achieve good effect on the control of nephrosclerosis,which may guide the clinical treatment of the renal sclerosis.
