摘 要:采用可见分光光度计测定蕤核叶总黄酮的含量,以总黄酮的吸附率、解析率为指标,比较分析AB-8、DM-130、HPD-100、HPD-600 4种树脂对蕤核叶总黄酮的分离纯化效果,确定最佳树脂型号及其最佳工艺条件。结果表明:AB-8对蕤核叶总黄酮吸附性能最好,吸附率为76.93%,解吸率为81.93%;最佳工艺条件为提取液pH为4.0,上样液的浓度为1 mg/mL^2 mg/mL,上样量与树脂的比例8∶1;洗脱剂选用60%乙醇,用量4 BV(柱体积)。蕤核叶总黄酮的含量在未纯化之前为15.3%,在最佳条件纯化之后可达到48.6%。The visible spectrophotometer to measure the content of total flavonoids brought Prinsepia uniflora Batal. leaves ,the adsorption rate and desorption rate of AB-8,DM-130,HPD-100,HPD-600 four kinds of macroporous resins were analysed to determine the suitable resin and its optimum process conditions. The results indicated:AB-8 resin was best absorbent for the total flavonoids in Prinsepia uniflora Batal.leaves, the adsorption rate was 76.93 % ,the desorption rate was 81.93 %.The optimum technical conditions were that the pH of extracts was 4.0, the concentration of sample solution was 1 mg/mL-2 mg/mL, the ratio of absorbing solution to resin was 8 : 1 ,the eluting solution was 60 % ethanol, the volume of the eluting solution was 4 BV. The content of total flavonoids brought Prinsepia uniflora leaves in not before purification for 15.3 %, in the best condition after purification can reach 48.6 %.
