摘  要:目的:建立RP—HPLC法同时测定芎菊上清丸中绿原酸、盐酸小檗碱和黄芩苷的含量。方法:采用DIKMA Platisil ODS色谱柱(250mm×4.6mm,5μm),柱温30℃,以甲醇(A)-0.1%磷酸水溶液(B)为流动相,梯度洗脱(0~5min,40%A;5~20min.40%A→60%A;20~30min,60%A),流速1mL·min^-1,变换波长检测(327nm,绿原酸;345nm,小檗碱;278nm,黄芩苷)。结果:绿原酸、盐酸小檗碱和黄芩苷的线性范围分别为0.11—2.2μg(r=0.9997)、0.021~0.42μg(r=0.9999)、0.03~0.6μg(r=0.9998),平均回收率在98.45%和99.07%之间。结论:该法检测效率高,专属性强,重复性好,可用于芎菊上清丸中绿原酸、盐酸小檗碱和黄芩苷的含量测定及其质量控制。Objective: To establish a RP - HPLC method for simultaneous determination of chlorogenic acid, berberine hydrocbloride and baicalin in Xiongju Shangqing pills. Methods: The separation was performed on a DIK- MA Platisil ODS column (250 mm×4.6mm,5μm) with gradient elution (0 -5 min ,40% A ;5 -20 min ,40% A→60% A ;20 -30 min ,60% A) using methanol(A) and 0.1% phosphoric acid aqueous solution(B) as mobile phase at a flow rate of 1 mL · min^-1. The detection wavelengths were 327 nm for chlorogenic acid ,345 nm for berberine and 278 nm for baicalin, respectively. Results: Linear ranges of chlorogenic acid, berberine hydrochloride and baicalin concentrations were 0.11 - 2.2 μg ( r = 0. 9997 ) , 0. 021 - 0.42 μg ( r = 0. 9999 ) , 0.03 - 0.6μg ( r = 0. 9998 ) ,respectively. Their average recoveries fell between 98.45% and 99.07%. Conclusion: The RP - HPLC method established can be used to determine the contents of chlorogenic acid,berberine hydrochloride and baicalin in Xiongju Shangqing pills,which may also be applied as a standard for the quality control of Xiongju Shangqing pills.
