消疮饮对兔耳微痤疮模型抑制作用及急性毒性实验研究

摘　　要：目的:评价消疮饮对兔耳微痤疮模型的作用,同时进行该药小鼠灌胃给药的急性毒性实验研究,确定其安全范围。方法:兔采用油酸涂抹及皮内注射表皮葡萄球菌的方法造模,分成6组,每组10只,为清热暗疮片组,异维A酸胶丸组,消疮饮低、中、高剂量组,正常对照及模型组,灌胃给药14 d后处死,观察其耳部皮肤组织学改变。另将50只小鼠平均分成5组,应用不同剂量的消疮饮浸膏灌胃给药,剂距为1∶0.9,测定小鼠灌胃给药最大耐受量及最小致死量。结果:消疮饮低、中、高剂量组的皮脂腺直径、毛囊面积均显著低于模型组。消疮饮浸膏对小鼠灌胃给药的最大耐受量和最小致死量分别为生药249.77 g/kg和生药277.53 g/kg,分别为成人临床拟用剂量的73.68倍和81.87倍(成人按60 kg体重计)。结论:消疮饮对兔耳微痤疮具有治疗作用,其临床服用剂量是安全的。Objective： To evaluate the effect of Xiaochuang Drink on rabbit ear micro acne model, at the same time, the rabbits were given experimental study on acute toxicity of the herb to determine the safe range. Methods ： Model rabbits induced by oleic acid coated and intradermal injection of Staphylococcus epidermidis molding, were divided into 6 groups, Qingre Anchuang tablets group, isotretinoin soft capsules group, Xiaochuang Drink low, middle, high dose groups, normal control and model group, 10 of each group. After 14 days of intragastric administration, the changes of its ear skin tissue were observed. The other 50 rabbits were divided into 5 groups, and different doses of Xiaochuang Drink extract were given by intragastric administration, with agent distance of 1 ： 0.9. The maximum tolerance and minimum lethal dose were measured. Results ： The sebaceous gland and hair follicle area diameter of Xiaochuang Drink low, middle and high dose groups were significantly lower than those in model group. The maximum tolerated dose of medicine and minimum lethal dose were 249.77g crude drug/kg and 277.53g crude drug/kg, respectively for the adult clinical dose of 73.68 times and 81.87 times adult 60 kg by weight ）. Conclusion ： Xiaochuang Drink has a therapeutic effect on rabbit ear micro acne, and the clinical dose is safe.
