摘  要:目的 :建立用反相高效液相色谱内标法梯度洗脱同时测定金银花中绿原酸、咖啡酸、 3,5 -二咖啡酰奎尼酸含量的方法 ,并对 4个产地 3种金银花中上述 3种化合物进行定量分析。方法 :采用岛津CLC -ODS柱 (6 0mm× 15 0mm) ,甲醇 - 10 %的磷酸氢二钠和醋酸缓冲液流动相不同比例进行梯度洗脱 ,内标物为水杨酸 ,检测波长 30 4nm。结果 :西南地区金银花药材主流商品灰毡毛忍冬、细毡毛忍冬干燥花蕾中绿原酸、咖啡酸、 3,5 -二咖啡酰奎尼酸含量高于山东、河南产金银花。结论 :本法灵敏快速 。Objective:A RP-HPLC method using an internal standard with gradient elution mode is developed for the separation and determination of chlorogenic acid,caffeic acid,3,5-O-dicaffeoylquinic acid in different resources from simultaneously.Methods:Experiments were performed on CLC-ODS column (6 0 mm×150 mm).The mobile phase used was a linear gradient of methanol in 10% Na 2HPO 4-HAc buffer.Salicylic acid was used as internal standard.The UV detector was set at 304 nm.Results:The results show that the contents of these compounds in Lonicera macrathoides Hands.-Mazz.and L.similis Hemsl.which are produced in the southwest region of China are higher than those in L. japonica Thunb. produced in Shandong province and Henan province.Conclusion:The method is sensitive,quick,with accurate and reliable results.
