摘 要:目的观察神经降压素在高血糖条件下对巨噬细胞的影响。方法取昆明小鼠脾巨噬细胞,高糖培养液培养15 d,细胞分PBS对照组和神经降压素干预组。ELISA检测法检测TNF-、IL-1β及IL-6的释放;RT-PCR检测TNF-、IL-1β及IL-6 mRNA的表达。结果高糖条件下神经降压素能降低巨噬细胞TNF-、IL-1β及IL-6的释放和mRNA的表达(P<0.05)。结论在高糖条件下,神经降压素可以通过抑制炎性细胞因子的释放影响巨噬细胞的反应。Objective To observe the effect of Neurotensin (NT) in macrophages function under hyperglycemic conditions. Methods Macrophages were prepared from spleen of Kunming mice and incubated in 30mM (high glucose) D-glucose, for 15 days. The ceils were divided into the PBS control group and Neurotensin treatment group. The level of TNF-α, IL-1β and IL-6 was detected by ELISA; The mRNA expressions of TNF-α, IL-1β and IL-6 were detected using RT-PCR. Results: NT can decrease the release of TNF-α, IL-1β and IL-6 in macrophages under hyperglycemic conditions (P〈0.05). Conclusion These studies demonstrate that NT through the regulation of inflammatory cytokine expression affects macrophage responses.
