机械拉伸对兔角膜成纤维细胞增殖和迁移的影响  被引量:2

Effects of Cyclic Mechanical Stretch on the Proliferation and Migration of Rabbit Corneal Fibroblasts

摘  要:对兔角膜成纤维细胞施加5%应变、0.1Hz的周期性机械拉伸,采用流式细胞仪检测细胞增殖情况,划痕法研究细胞迁移情况。结果发现,与静态对照组相比,5%机械拉伸能够使S期细胞数量明显增多;施加1ng/mL表皮生长因子(Epidermal growth factor,EGF)或100ng/mL碱性成纤维细胞生长因子(Basic fibroblast growth factor,bFGF)后细胞相对划痕宽度明显变小。与各自静态对照组相比,拉伸10h,单轴拉伸组相对划痕宽度明显增加,等双轴拉伸则无显著性差异;拉伸20h,无论是等双轴还是单轴拉伸,相对划痕宽度均明显增加;拉伸同时施加bFGF或EGF后相对划痕宽度均减小,与静态对照组相当。提示机械拉伸能够促进角膜成纤维细胞增殖,但抑制了细胞迁移行为,该抑制效应与拉伸作用方式有关;而bFGF和EGF则可以减弱牵拉引起的细胞迁移抑制效应。In this study, isolated rabbit corneal fibroblasts were subjected to the regimen of 5% stretch at 0. 1 Hz. Cell proliferation was detected using flow cytometry. Multiple wounds were scraped through monolayer cells to investigate cell migration. Our results showed that 5 % stretch significantly increased the percent of S phase in the total cell cycle. Relative wound width became smaller when cells were treated with 1 ng/mL epidermal growth factor (EGF) or 100ng/ mL basic fibroblast growth factor (bFGF). After 10 h stretch, relative wound width of uniaxial stretch groups became larger than the unstretched control. No significant differences were found in the equibiaxial stretch groups. After 20 h, whether equibiaxial or uniaxial stretch, relative wound width was obviously larger than that for unstretched control. The relative wound width decreased when cells were treated with both bFGF or EGF under cyclic stretch, and was close to that of unstretched control. It suggests that stretch promotes the proliferation of the corneal fibroblasts, but inhibits the cell migration behavior, this inhibitory effect can be attenuated by bF- GF or EGF, and is associated with the applying manner of the mechanical force.
