摘  要:目的:观察深低温对大鼠脊髓缺血再灌注的保护作用及对Caspase12表达的影响,初步探讨其可能的作用机制。方法:将45只SD大鼠随机分为3组:深低温18℃处理组(A组);常温手术对照组(B组);常温假手术组(C组)。每组分为6h、24h、1周三个不同时间点取L2~L5段的脊髓,每个时间点5只大鼠,采用普通病理HE染色、Nissl染色、电镜从形态学上观察脊髓组织,免疫组化检测和RT—qPCR检测缺血再灌注后脊髓组织Caspase12蛋白和mRNA的表达变化。结果:组织形态学观察A、B组病理改变均较c组严重,且A组比B组轻.病理学累积评分A、c组明显低于B组(P〈0.01);免疫组化检测和RT-qPCR检测结果分别显示,Caspase12蛋白和mRNA在A、B组各时间段均高于C组,且A组升高程度比B组低(P〈0.05)。结论:深低温对大鼠脊髓缺血再灌注有保护作用。并且可能通过抑制Caspase12的表达而发挥其保护作用。Objective To evaluate the effects of deep hypothermia on spinal cord ischemia reperfusion injury (SCII) and on the expression of Caspasel2 in spinal cord in rats. Methods 45 rats were divided into 3 groups: 18℃ administration group (Group A, n = 15), 37℃ administration group (Group B, n = 15), and sham operation control group (Group C, n = 15). L2-5 spinal samples were collected at 6 h, 24 h, and 1 week after the administration from rats (each n = 5) in the 3 groups; and then the expressions of Caspasel2 mRNA and protein were detected by RT-qPCR and immunohistochemistry, respectively; the histological changes of spinal cord were observed by HE staining, Nissl staining under light and electron microscopy. Results The pathological changes in Group A and B were worse than those in Group C, and were the worst in Group B (P 〈 0.01). The expressions of Caspasel2 mRNA and protein in Group A and B at the three time points were significantly higher than those in Group C, and were the highest in Group (P 〈 0.05). Conclusions Deep hypothermia may protect the spinal cord from SCII through inhibiting the expression of Caspase 12.
