摘 要:[目的]探索一种原位沙培内生根的根呼吸测定方法,并同离体测定方法相结合观测成熟枇杷[EHobotryaJaponica(Thunb.)Lindl.]果树的细根呼吸。[方法]将枇杷细根放入装有洗脱了有机质河沙的根室内培养11d后,分别在5个时间点每隔2h用Li-8100CO:测定系统(Licor,USA)测定内生根的CO:排放量。[结果]所有内生根样品直径范围在0.69~2.15mm,非内生根样品直径范围在0.66~1.70mm;根呼吸速率在近地表温度20.5~30.5℃和0~5cm土层温度15.9~23.3℃范围内测定的平均值为7.52±4.96nmol/(DM^2·s),非内生根离体测定值1.76±0.20nmol/(DM^2·S),内生根法原位测定值是非内生根离体测定值的3倍。[结论]内生根方法通过改进可以用于成熟林木细根呼吸乃至其他代谢活性研究。[ Objective ] A root in-growth approach was explored to measure respiration of forest tree roots in situ and combined with detached root method root respiration of loquat trees (Eriobotrya Japonica ( Thunb. ) IAndl. ) was measured. [ Method ] Loquat roots were grown into chambers filled with sand washed off organic matter for 11 days and respiration rates were measured with Li-8100 system every 2 hours for 5 dates. [ Result ] Diameters of the roots in in-growth chambers and out of chambers ranged from O. 69 -2.15 mm and 0.66 - 1.70 mm, respectively. Over near-surface temperature range of 20.5 - 30.5 ℃ and 0 - 5 cm depth soil temperature of 15.9 - 23.3℃, respiration rates measured'averaged as 7.52 ± 4.96, 1.76 ± 0.20 nmol/( DM2 . s), respectively. Respiration rates for in-growth roots were 3 times higher than those for detached roots outside chambers, respectively, concurring with literature observations. [ Conclusion ] The root in-growth approach, after further improve- ment, appears promising for providing information for the metabolic activity of fine roots of mature forest trees growing in soil.
