摘 要:目的探讨白花丹参提取物对脂多糖损伤内皮祖细胞周期与凋亡的作用。方法以10μg/mL脂多糖诱导内皮祖细胞损伤,制备内皮祖细胞损伤模型后用不同质量浓度白花丹参提取物干预,用MTT比色法检测各组细胞活性,用AnnexinV/PI双染法检测细胞凋亡变化,碘化丙啶单染流式细胞仪检测各组细胞周期。结果与正常对照组比较,模型组细胞增殖活性明显下降,细胞凋亡率明显增加,S期细胞比率降低,G0/G1期细胞比率增加。与模型组比较,白花丹参组细胞存活率升高,细胞凋亡率下降,且呈剂量依赖性,G0/G1期细胞比率降低,S期及G2/M期细胞比率均增加。结论白花丹参对脂多糖损伤的血管内皮祖细胞有保护作用,其机制可能是通过保护细胞周期正常进行、抑制细胞凋亡实现的。AIM To explore the effect of Salvia miltiorrhiza Bunge. f. alba extract on the cell cycle and apoptosis of endothelial progenitor cells injured by lipopolysaccharide (LPS). METHOD The injury to endothelial progenitor cells was induced by 10μg/mL LPS and Salvia miltiorrhiza Bunge. f. alba extract had been intervening in endothelial progenitor cells. Changes of cellular morphology were detected with microscope. The viability of the cells in each group was examined by MTT method. Flow cytometry Annexin V/PI staining was used for apoptotic changes and PI staining for analysis of the effect of the cell cycle. RESULTS Compared with the control group, LPS decreased the proliferative activity and increased the rate of cell apoptosis. Flow cytometry PI staining showed there were more cells arrested at G0/G1 stage and less cells at S stage in LPS damage group, rather the other way around in Salvia miltiorrhiza Bunge. f. alba extract group. CONCLUSION Salvia miltiorrhiza Bunge. f. alba extract may have significant protective effect on the endothelial progenitor cells injured by LPS and related to the protection of normal cell cycle and the decrease in the cell apoptosis.
