高效液相色谱法同时测定一清颗粒中3种原小檗碱型生物碱的含量

摘　　要：目的:建立高效液相色谱(RP-HPLC)法同时测定一清颗粒中原小檗碱型生物碱的含量。方法:采用DIONEX C18(4.6 mm×250 mm,5μm)色谱柱;流动相为乙腈-0.05 mol·L-1磷酸二氢钾溶液(用磷酸调节pH值至3.0)(30∶70),流速1.0ml·min-1,检测波长350 nm。结果:小檗碱、黄连碱、巴马汀3种原小檗碱型生物碱在本文条件下,分离良好;标准曲线的相关系数(r)分别为1.0000、0.999 9、0.999 9;平均回收率(n=9)分别为101.2%,101.8%,101.7%,RSD分别为0.95%,0.48%,0.73%。结论:本方法准确,重复性良好,简单易行,适用于同时测定一清颗粒中3种原小檗碱型生物碱的含量。OBJECTIVE To establish a quantitative method for analyzing the content of berberine-type alkaloids in Yiqing granules by RP-HPLC.METHODS The separation was carried on DIONEX C18Column（Ф4.6 mm×250 mm,5μm）.The mobile phase was acetonitrile：0.05 mol·L-1 potassium dihydrogen phosphate（Using phosphoric acid adjusted PH to 3.0）（30∶ 70）,The flow rate was 1.0ml·min-1,and the detection wavelength was set at 350 nm.RESULTS 3 berberine-type alkaloids could be separated well.All the correlation coefficients were 0.999 9 or above.The average recoveries（n=9）were 101.2%（RSD=0.95%）,101.8%（RSD=0.48%）,101.7%（RSD=0.73%）.CONCLUSION The method described above is accurate, simple,and reproducible,can be used to determine 3 berberine-type alkaloids in Yiqing granules at the same time.
