摘 要:目的:探讨过氧化氧化还原蛋白(PRDX)ⅠmRNA在小鼠卵泡发育、排卵及卵巢衰老过程中的重要作用。方法:分别选用不同卵巢功能(形成期、性未成熟期、性成熟期、功能退化期)和不同动情周期(动情前期、动情期、动情后期、动情间期)以及促性腺激素预处理不同时间点[注射0.9%氯化钠液(CON)后、注射孕马血清(PMSG)48小时(P48h)后和注射绒促性素(HCG)后4小时(H4h)至48小时(H48h)]C57BL/6j小鼠的卵巢,通过实时定量聚合酶链反应检测PRDXⅠmRNA在卵巢中的表达。结果:①PRDXⅠmRNA在性未成熟期组、性成熟期组和功能退化期组小鼠卵巢中稳定表达,组间比较,差异无统计学意义(P>0.05),但均较形成期组低,差异均有统计学意义(P<0.05)。②在性成熟小鼠不同动情周期间表达比较,差异无统计学意义(P>0.05)。③在促性腺激素预处理卵巢中,与CON组相比,PRDXⅠmRNA在PMSG作用后48小时(P48h组)表达显著升高(P<0.05),而在HCG作用后8小时(H8h组)最低(P<0.05),后于48小时(H48h组)升高(P<0.05)。结论:PRDXⅠmRNA在卵泡发育、排卵和卵巢衰老中可能发挥调控作用。Objective: Exploring the expression of peroxiredoxin I mRNA in the different stages of C57BL/6j mouse follicle development,ovulation and ovarian aging. Methods:Real-time polymerase chain re- action was used to detect the expression of PRDX I mRNA in different stages of ovarian age(formation,im- mature, maturation and degeneration stage ), estrous ( p^oestrus, est^us, metaestrus, diestrus) and gonadotro- phin-primed immature C57BI_/6j mouse ovariesE NS injection as a control group(CON) ,48 hours after serum gonadotrophin injection as the P48h group and 4 to 48 hours after human chorionic gonadotrophin injection as the H4h to H48h group]. Results :~)ln the ovaries of different age stage of C57BL/6j mice,we found that there was no difference of the PRDX I mRNA expression the immature stage,maturation stage and degen- eration stage( P〉O. 05) ,but all lower than that in the formation stage( P 〈0.05). ~ There were no differ- ence of the PRDX I mRNA among estrous cycles( P〉0. 05). We noticed that in the ovaries of the gona- dotrophin primed immature mouse the expression of PRDX I mRNA increased 48 hours after PMSG injection ( P〈O. 05) ,and diminished to the lowest level 8 hours after HCG treatment t( P〈O. 05) ,and then increased at 48 hours( P 〈 0. 05). Conclusions:The PRDX IV probably plays important roles in the follicle development and ovarian aging.
