摘 要:目的探讨杀伤性免疫球蛋白样受体(KIR)在丙型肝炎病毒(HCV)感染人群中基因多态性分布,为研究KIR在HCV感染中的作用机制奠定基础。方法从广州血液中心2009年2月至2012年6月采集的HCV抗体检测呈阳性的无偿献血者标本中,计算机随机数表法随机抽取170份无血缘关系的样本作为研究对象。对其采用聚合酶链反应一序列特异的寡核苷酸扩增(PCR-SSO)技术检测KIR基因表型,并计算其基因频率。结果KIR2DL4、3DL2、3DL3、3DPI存在于所有样本,其基因频率为1.00。KIR2DL1、2DP1、2DL3、2DS4、3DLl频率较高,分别为0.923,0.923,0.923,0.684,0.770;KIR2DL2、2DL5、2DSl、2DS2、2DS3、2DS5、3DSl基因频率较低,分别为0.129,0.244,0.214,0.119,0.116,0.139,0.218。共发现30种KIR基因型,其中1型最为常见,频率为0.459。另外,还发现3种新的基因型。与报道的中国健康人KIR基因相比,KIR2DS3的频率较高,差异具有统计学意义(X^2=4.733,P=0.030)。结论广东地区HCV感染献血人群具有其独特的KIR基因表达频率、单倍型和基因型频率分布。KIR2DS3基因可能参与了HCV的感染过程。Objective To investigate the distribution of killer immunoglobulin-like receptor (KIR) genes among hepatitis C virus (HCV) infected blood donors in Guangdong province and provide basic data for further research of mechanism between KIR and HCV infection. Methods From February 2009 to June 2012, a total of 170 HCV infected individuals were randomly selected from volunteer blood donors in Guangdong by computer. The genotypes of KIR were detected by polymerase chin reaction restriction sequence specific oligonucleotide (PCR-SSO), followed by frequency calculating and statistical analysis. Results Among the Guangdong population of HCV infection, framework genes KIR 2DL4, 3DL2, 3DL3 and 3DP1 were found in all samples. Genes KIR 2DL1, 2DP1, 2DL3, 2DS4 and 3DL1 were in high frequencies with 0. 923, 0. 923, 0. 923, 0. 684 and 0. 770 respectively. Genes KIR 2DL2, 2DL5, 2DS1, 2DS2, 2DS3, 2DS5, 3DS1 were in low frequencies with 0.429, 0. 244, 0. 214, 0. 119, 0. 116, 0. 139 and 0. 218 respectively. 30 genotypes were observed, among which genotype 1 was the most common with the gene frequency of 0. 459. Three new genotypes were identified in this research. KIR2DS3 had a higher frequency in HCV infected individuals than in healthy populations in China (2 ~ 4. 733, P ~ 0. 030). Conclusions These findings suggest that there are distinctive frequencies of KIR genes content, haplotype as well as genotype in Guangdong population of HCV infection. KIR2DS3 gene may be involved in HCV infection. Our study might provide valuable evidence for the research of the correlation between KIR and HCV infection.
