摘 要:[目的]评价异种骨与成骨细胞体外生物相容性,为其在临床应用提供实验依据。[方法]将SD大鼠来源的成骨细胞分别在普通培养基和含异种骨浸提液的培养基中培养,于1、3、5、7 d用四甲基偶氮唑盐(methyl thiazolyltetrazolium,MTT)法测定吸光度值,以评价细胞的增殖情况。采用Transwell试验,在倒置相差显微镜下观察细胞的迁移情况,计算穿膜成骨细胞数量,检测成骨细胞的迁移能力。应用流式细胞仪检测细胞周期变化,以评价异种骨浸提液对成骨细胞周期的影响。[结果]成骨细胞在普通培养基和含异种骨浸提液的培养基中采用MTT检测增殖结果无显著性意义(P>0.05)。在含异种骨浸提液的培养基中,成骨细胞在Transwell中穿膜细胞数为(13.40±2.51)个/视野,对照组为(8.00±1.87)个/视野,两组差异有显著性意义(P<0.05),实验组细胞迁移数量多于对照组。流式细胞仪检测显示,异种骨浸提液对细胞周期无影响。[结论]异种骨与成骨细胞有良好的生物相容性,为其临床应用提供了试验依据。[ Objective] To access the biocompatibility of xenogeneic bone and osteoblasts in vitro, so as to provide the ex-perimental data for clinical application. [ Method ] The SD rat osteoblasts were cultured in mediums with or without extracts from xenogeneic bone, and the proliferation of osteoblasts was quantitively evaluated by 3 - [ 4,5 -dimethylthiazol -2 -yl ] -2,5 -diphenyl tetrazolium bromide (MTT) colorimetly method at intervals of 1,3,5,7 d. Cell migration was observed by inverted phase contrast microscope by transwell assay, the cell cycle was detected by flow cytometry. [ Result ] MTT study showed that ex-tracts from xenogeneic bone had no statistical difference for the growth of osteoblasts contrast ( without extracts) ( P 〉 0.05 ). Transmembrane cell number was 13.40 ± 2.51 cells/field in the experiment group compared with 8.00 ± 1.87cells/field in the control group, there was significant difference, nmnber of cell migration in the experiment group was more than in the control group. Flow cytometry showed that extracts from xenogeneic bone had no effect on the cell cycle. [ Conclusion] The xenogeneic bone has good cytocompatibility with osteoblast,it provids experimental data for its clinical application.
