摘  要:目的比较改良M199培养基与进口M199培养基培养Vero细胞及狂犬病病毒的效果,旨在筛选出更适合Vero细胞生长及其相关病毒培养的培养基。方法分别用含5%新生牛血清的改良M199培养基和进口M199培养基培养Vero细胞,于倒置显微镜下观察细胞的生长状态,并进行细胞计数,台盼蓝染色法测定细胞活力;分别用含1%新生牛生血清的改良M199培养基和进口M199培养基培养狂犬病病毒,采用小鼠脑内滴定法测定病毒滴度。结果改良M199培养基培养的Vero细胞形态更规则,增殖更快,细胞密度和活力均高于进口M199培养基培养的Vero细胞;使用改良M199培养基培养狂犬病病毒,病毒增殖更快,滴度更高,峰值可达109log LD50/ml,明显高于使用进口M199培养基培养的病毒滴度。结论改良M199培养基更适合Vero细胞以及狂犬病病毒的培养。Objective To compare the applicability modified and imported M199 media in culture of Vero cells and ra- bies virus so as to screen more suitable medium for growth of Vero cells and culture of related viruses. Methods Vero cells were cultured by modified and imported M199 media, each containing 5% newborn calf serum, respectively, then observed for morphology and counted under inverted microscope, and determined for activity by trypan blue staining. Ra- bies virus was cultured by modified and imported M199 media, eaeh containing 5% newborn calf serum, respectively, and determined for titer by intracerebral titration in mice. Results The Vero cells cultured in modified M199 medium were more regular in shape and grew rapidly, of which the density and activity were higher than those in imported M199 medium. However, the rabies virus cultured in modified M199 medium was proliferated rapidly as compared with that in imported M199 medium, and the titer of the former was significantly higher than that of the latter, with a peak value of 109 lgLD50/ml. Conclusion Modified M199 medium was more suitable for cultures of Vero cells and rabies virus.
