二尖瓣狭窄合并心房颤动患者HCN2表达与测序

摘　　要：目的:探讨超级化激活环核苷酸调控通道基因家族-2(HCN2)基因在风湿性心脏病二尖瓣狭窄合并心房颤动患者右心耳组织中的表达。方法:采用实时荧光定量聚合酶链反应(FQ-PCR)的方法分析HCN2mRNA在43例风湿性二尖瓣狭窄患者右心耳组织中的表达,并对反应产物进行测序。结果:测定HCN2基因cDNA序列同源性为100%,风湿性心脏病二尖瓣狭窄合并心房颤动组HCN2mRNA表达量明显高于窦性心律组(P<0.01),HCN2mRNA的表达与左心房内径、心胸比例呈正相关,而与二尖瓣瓣口面积呈负相关。结论:风湿性心脏病二尖瓣狭窄患者并发心房颤动的发生发展过程中,HCN2mRNA表达明显上调。Objective：To explore the expressions of HCN2in atrial fibrillation patients with mitral stenosis of rheumatic heart disease（RHD）.Method：The expression of HCN2in 43mitral valve stenosis of RHD were detected with real-time fluorescence quantitative-PCR（FQ-PCR）,and the PCR products of HCN2gene sequences were analyzed.Result：The homology of HCN2gene cDNA sequence was 100%,and the expressions of HCN2mRNA in atrial fibrillation patients with mitral stenosis of RHD were obviously higher than those in control group（P 0.01）.The mRNA of HCN2was not only significantly positive correlated with LAD and CTR,but also negatively correlated with the area of mitral valve.Conclusion：The up-regulated expressions of HCN2plays an important role in the atrial fibrillation of RHD.
