摘 要:目的研究900 MHz电磁辐照对大鼠学习记忆能力、血脑屏障通透性和脑组织超微结构的影响。方法 48只健康成年雄性SD大鼠分为辐照14 d组(EMF 14 d组)、辐照28 d组(EMF 28 d组)、假辐照14 d组(Sham 14 d组)和假辐照28 d组(Sham 28 d组)4组,每组12只。辐照组分别连续暴露于900 MHz电磁波(功率密度为1 mW/cm2)14 d和28 d,3 h/d。Morris水迷宫观察大鼠神经行为学,血清白蛋白免疫组化观察海马神经元受损情况和反映血脑屏障通透性变化,透射电镜观察组织超微结构。结果 Morris水迷宫实验结果显示EMF 28 d组通过目标象限百分比和穿越平台次数明显少于其他组(P<0.05)。900 MHz电磁辐照后,与假辐照组(0 mW/cm2)相比,辐照组(1 mW/cm2)血清白蛋白在脑内高表达(P<0.05)。透射电镜结果显示辐照组较多神经元损伤,血脑屏障结构破坏。结论 900 MHz电磁辐照可致大鼠学习记忆能力下降,增加血脑屏障通透性,造成海马神经元的损伤,其与微波辐照的时间可能有一定的关系。Objective To investigate the effects of low-intensity microwave (900 MHz) electro- magnetic field (EMF) on the spatial memory, permeability of blood-brain barrier and uhrastructural changes of related rat brain regions. Methods Male SD rats were randomly divided into 4 groups (n = 12 for each group) and separately underwent EMF exposure for 14 d (EMF 14 d group), EMF exposure for 28 d (EMF 28 d group), sham-exposure for 14 d (sham 14 d group) and sham-exposure for 28 d (sham 28 d group). Continuous microwave EMF exposure (900 MHz, power density = 1 mW/cm2) was conducted to the head of rats for 3 h per day in the EMF groups. Then, behavioral changes were examined using the Morris water maze. The permeability of the blood-brain barrier and signs of neuronal damage in hippocampal region were assessed by immunohistochemical staining for albumin. The ultrastructural changes of the brain tissue were observed by transmission electron microscopy (TEM). Results Compared with the sham exposure groups (0 mW/cm2), the EMF exposure groups (1 mW/cm2) showed obviously changed intracellular ultrastructure of brain tissue, damaged structure of blood-brain barrier, and significant increased albumin extravasation and uptake into neurons (P 〈 0.05 ). The percentage of target quadrant and the times of crossing platform were significantly decreased ( P 〈 0.05 ) in the EMF 28 d group. Conclusion Exposure to 900 MHz microwaves ( 1 mW/cm2 ) may alter the ability of spatial memory, change blood-brain barrier permeability, and induce the uhrastructural changes in rat hippocampal neurons.
