摘 要:目的探讨拉贝洛尔对创伤性脑损伤(TBI)大鼠空肠通透性的影响。方法 54只雄性Wister大鼠,随机分为对照组(n=18)、TBI组(n=18)、TBI+拉贝洛尔组(n=18);每组伤后3、6、12h时间点各分为3个亚组(n=6),参照Feeney’s方法建立大鼠TBI模型。在各时间点采用异硫氰酸荧光素葡聚糖(FITCDextran)行大鼠活体内空肠通透性评价,酶联免疫吸附剂测定法(ELISA)测定血浆肾上腺素、肠道肿瘤坏死因子-α(TNF-α)水平,光学显微镜下观察各组肠黏膜组织形态变化。结果 TBI组大鼠在建模后各时间点血浆肾上腺素水平、肠道TNF-α水平和肠道通透性较对照组明显增加(P<0.01),TBI+拉贝洛尔组大鼠血浆肾上腺素水平和肠道TNF-α水平在建模后各时间点较TBI组大鼠明显降低(P<0.01,P<0.05),建模后6、12h TBI+拉贝洛尔组大鼠的肠道通透性较TBI组降低(P<0.05)。结论拉贝洛尔能降低大鼠TBI后空肠通透性的增加。Objective To study the effect of labetalol on tumor necrosis factor - alpha ( TNF - ct ) and per meability in rats after traumatic brain injury (TB[). Methods Fifty-four male Wister rats were randomly divided into three groups : control group ( n = 18 ) , TBI group { n = l 8 ) , TBI + labetalol group ( n = 18 ). Each group was fur-ther divided into 3 hours ,6 hours and 12 hours subgroups ( n = 6/subgroup ). The rat TBI model was established by referring to Feeney' s method. After the model development, animals underwent assays of vivo intestinal permeabili- ty,evaluation of plasma epinephrine and intestinal TNF-aby ELISA method at each time point. And the pathologi- cal changes of intestinal mucosa were observed under light microscope. Results The levels of plasma epinephrine, intestinal TNF-a and permeability at each time point in TBI group were significantly higher than those in control group { P 〈 0. 01 ). In TBI + labetalol group, the levels of plasma epinephrine and intestinal TNF-a at each time point were significantly lower than those in TBI group ( P 〈 0. 01, P 〈 0. 05 ). At 6 and 12 hours after model establish- ment, the level of intestinal permeability in TBI + labetalol group was significantly lower than that in TBI group ( P 〈 0. 05 }. Conclusion Labetalol can decrease the levels of intestinal TNF-a and lessen the increase of intestinal per- meability of rats in TBI model.
