摘 要:目的:探究脊髓损伤(spinal cord injury,SCI)后应用甲基强的松龙(methylprednisolone,MP)治疗对损伤脊髓节段中的炎性细胞因子IL-6、IL-10和自噬因子LC3B表达的影响。方法:采用改良Allen氏法建立大鼠脊髓(T10)损伤模型,60只SD雌性大鼠随机分为假手术组(n=20)、手术组(n=20)和MP组(n=20)。MP组于SCI后立刻经尾静脉给予MP(30 mg/kg),假手术组只打开椎板不损伤脊髓。分别于损伤后4 h、12 h、1 d、3 d和7 d取材,采用半定量RT-PCR方法检测损伤段脊髓组织中炎症因子IL-6、IL-10 mRNA和自噬因子LC3B mRNA的表达变化。结果:假手术组动物脊髓组织中IL-6、IL-10、LC3B mRNA的表达微弱。手术组的IL-6、IL-10、LC3B mRNA的表达均在损伤后4 h开始明显升高,于损伤后1 d达最高峰值,之后逐渐降低(P﹤0.05)。手术组的LC3B与IL-6和IL-10的表达呈现相似的趋势,IL-6、IL-10均与LC3B呈正相关。MP组的IL-6、IL-10、LC3B mRNA的表达在损伤后12 h开始明显受到抑制,到1 d时达最低值,7 d时LC3B mRNA的表达仍低于手术组(P﹤0.05),MP组的LC3B与IL-6和IL-10的表达也是呈现相似的趋势,IL-6、IL-10也均与LC3B呈正相关。结论:大鼠脊髓损伤后应用甲基强的松龙可以通过抑制炎症而抑制损伤节段的自噬活动。Objective: To investigate the effect of methylprednisolone (MP) on the expression of inflammatory cytokines IL-6, IL-10 and autophagy factor LC3B in injured spinal cord segment of rats after spinal cord injury. Methods: The model of spinal cord injury(SCI) was established by using the modified Allen's method. 60 female SD rats were divided into three groups : Sham operation group ( n = 20), operation group ( n = 20 ) and MP group ( n -- 20 ). The rats in MP group was given by tail intravenous injection (30 mg/kg) after SCI, and those rats only opened laminectomy without dam- aging the spinal cord in sham operation group. The spinal cord tissues were removed at 4 h, 12 h, 1~t day, 3rd day and 7th day after operation. The expression change of the inflammatory cytokines IL-6, IL-10 and autophagy factor LC3B in spinal cord tissue was detected by using semi quantitative RT- PCR method. Results: The expression of IL-6, IL-10 and LC3B mRNA was lower in the shame group, whereas the expression of those mRNA was increased significantly in the operation group at 4 h after operation, and reached the peak at 1st day after operation, then gradually reduced ( P 〈 0.05 ). The trend of the expression of LC3B, IL-6 and IL-10 was similar in operation group. The LC3B was respectively positive cor-related with IL-6, IL-10 in operation group. The expression of IL-6, IL-10 and LC3B in MP group was inhibited from 12 h after operation, reaching the minimum at 1st day after operation. The expressions of LC3B were lower than those in op- eration group at 7'h day after operation( P 〈 0.05 ). The trend of the expression of LC3B and IL-6, IL-10 was similar in MP group. The expression of LC3B was also respectively positive correlated with IL-6, IL-10 in MP group. Conclusion: Methylprednisolone can inhibit the autophagy through inhibiting the inflammatory response in the segment of injured spinal Cold.
