富硒绿茶对大鼠非酒精性脂肪肝拮抗作用研究

摘　　要：目的探讨富硒绿茶对非酒精性脂肪肝的拮抗作用。方法将32只Wistar大鼠分成4组,分别饲喂标准饲料+常规饮水(对照组,NC)、高脂饲料+常规饮水(高脂模型组,HC)、高脂饲料+普通绿茶(普通绿茶组,T)、高脂饲料+富硒绿茶(富硒绿茶组,SeC),7w后处死,用苏丹Ⅲ染色肝脏组织切片观察脂肪化程度,用免疫组织化学方法测定肝脏细胞色素P450ⅡE1表达情况。并分析其肝功能、血脂及肝组织中谷胱甘肽过氧化物酶(GSH-Px)和丙二醛(MDA)的变化。结果 HC组在肝脏脂肪化程度、细胞色素P450ⅡE1(CYP450ⅡE1)表达情况都明显高于T、SeC、NC组,且SeC组低于T组。HC组血清TC、TG升高,T组、SeC组较HC组显著降低(P<0.01)。HC组肝组织中MDA显著升高;T组、SeC组肝组织中MDA较HC组降低(P<0.01),但MDA值仍较NC组高。HC组GSH-Px显著降低,T组、SeC组较HC组有显著升高(P<0.01)。结论富硒绿茶能有效降低高脂饮食大鼠肝脏脂肪变性程度、降低肝脂肪细胞色素P450ⅡE1表达,富硒绿茶对高脂饮食大鼠非酒精性脂肪肝具有拮抗作用。Objective To explore the antagonistic action of selenium-enriched green tea on non-alcoholic fatty liver in rats. Methods Thirty two Wistar rats were divided into 4 groups randomly and fed with standard diet and water （NC）, high-fat diet and water （HC）, high-fat diet and tea （T）, high-fat diet and selenium-enriched tea （SeC） respectively. After 7 w, the rats were killed to investigate the pimelosis level in liver tissues by Sudan Ⅲ staining and the expression of hepatic cytochrome P450ⅡE1 by immunohistochemically. We also analyzed the changes of liver function, blood lipids, the glutathione peroxidase （GSH-Px） and malonyldialdehyde （MDA） in liver. Results The pimelosis level and the expression of P450Ⅱ E1 in HC group were significantly higher than those in T, SeC, NC groups. The levels of TC, TG and MDA in HC group were elevated. However, they were significantly down-regulated in intervention group compared to that in HC group （P〈0.01）. On the contrary, the level of GSH-Px in HC group was largely reduced and markedly up-regulated in intervention group （P〈0.01）. Conclusion Selenium-enriched tea effectively decreased the pimelosis level and P450 Ⅱ E1 expression in rats fed with high-fat diet and protected against the progression of non- alcoholic fatty liver.
