摘 要:目的观察马齿苋总黄酮对过氧化氢诱导人血管内皮细胞氧化损伤的保护作用。方法采用体外培养人脐静脉内皮细胞(HUVEC),实验分为5组:正常对照组、过氧化氢损伤组、马齿苋总黄酮低、中、高剂量组。MTT法测定各组细胞活力,荧光分光光度计测量各组细胞线粒体膜电位水平。结果与正常对照组比较,过氧化氢损伤组血管内皮细胞活力、线粒体膜电位显著降低(P<0.01);与过氧化氢损伤组比较,马齿苋总黄酮干预组血管内皮细胞活力、线粒体膜Ratio值明显升高(P<0.05),且存在明显的剂量依赖关系。结论马齿苋总黄酮对氧化应激诱导血管内皮细胞损伤具有一定的保护作用,其机制可能与其抗氧化、维持线粒体膜电位有关。Objective To explore the protective effects of portalaca total flavone(FIT) on human vascular en- dothelial cell (HUVEC) injury induced by oxidative hydrogen in vitro. Methods Human vascular endothelial cells was cuhured in vitro and divided into 5 groups: control group, hydrogen peroxide injury group, PTF low, medium and high dose group. Cellular vitality of all groups was determined by MTF method, level of mitochondrial mem- brane potential measured by fluorescence spectmphotometer. Results Compared with control group, cellular vitality and mitochondrial membrane potential of hydrogen peroxide injury group decreased significantly ( P 〈 0.01 ). Compared with hydrogen peroxide injury group, cellular vitality and mitochondrial membrane potential of portulaca flavonoids intervention group was significantly higher ( P 〈 0.05 ), and with obviously dose - dependent relation- ship. Conclusion Portalaca total flavone(PTF)has certain protective effect on human vascular endothelial cell in- jury induced by oxidative hydrogen in vitro and its mechanism may be related to its antioxidant, maintain mito- ehondrial membrane potential.
