摘 要:为探明青海地区引起奶牛乳房炎的优势病原并为主要致病菌的筛选和快速分型提供方法,用生化试验和16SrRNA保守序列PCR方法鉴定分离的金黄色葡萄球菌,用ERIC-PCR和RAPD方法进行基因分型,应用SPSS 17.0统计分析软件对两种分型扩增图谱进行聚类分析。结果共分离鉴定出金黄色葡萄球菌21株。用ERIC-PCR分型方法将21株金黄色葡萄球菌分为4个聚类群,其中Ⅰ型占42.86%(9/21),Ⅲ型占47.62%(10/21),Ⅱ型占4.76%(1/21),Ⅳ型占4.76%(1/21)。用RAPD分型方法将21株金黄色葡萄球菌分为3个聚类群,其中,A和C类群均有9株,各占42.86%;B类群仅有3株,占14.29%。分析发现,两种基因分型方法的结果基本一致,A、C两个类群的菌株数占总分离数的85.71%,为该地区金黄色葡萄球菌流行的优势菌群。说明引起该区奶牛乳房炎的金黄色葡萄球菌基因型有一定的相似性。In order to understand the incidence of dairy cow mastitis in Qinghai area caused by Staphylococ- cus aureus ,and provided a method of fast typing and screening major pathogens from dairy cow mastitis, S. aureus strains were isolated and characterized by biochemical analysis and 16 S rRNA sequence. ERIC- PCR and RAPD methods were used for genotyping,and the dendrogram and cluster were statistically ana- lyzed with SPSS 17.0 to evaluate the genetic similarity of the isolated S. aureus from this area. 21strains of S. aureus were isolated from the samples. And the 21 strains of S. aureus would also be catergoried into four group by ERIC-PCR cluster analysis,9 strains in type I (42.86%) ,10 strains in type [lI (47.62%) ,1 strain in type ]I (4.76 %), 1 strain in type 1V (4.76 %) ; When using the RAPD grouping system, all the 21 strains are catergoried into three groups,type A 9 stains (42.86%) and type C 9 strains(42.86%),while other 3 strains are type B (14.29%). The predominant genotypes of S. aureus strains in this area are types A and C, these strains accounted for 85.71% of the total . The study results showed that distributions of genotypes causing mastitis in dairy cows have a certain similarity in the region.
