摘 要:目的:建立同时测定维血宁中虎杖苷、大黄素-8-O-β-D葡萄糖苷及大黄素含量的方法。方法:应用HPLC法测定,色谱柱:Kromasil C18Column(4.6 mm×250 mm,5μm);流动相:乙腈-0.1%磷酸溶液,梯度洗脱;流速:1.0 mL·min-1;柱温:30℃;检测波长:288 nm。结果:虎杖苷、大黄素-8-O-β-D葡萄糖苷和大黄素的线性范围分别为0.09-0.72μg(r=0.9984)、0.4825-3.86μg(r=1.0000)、0.2675-2.14μg(r=1.0000);平均加样回收率分别为98.9%(RSD%=2.8%)、99.6%(RSD%=0.16%)、98.1%(RSD%=0.44%)。结论:该方法同时测定维血宁中的虎杖苷、大黄素-8-O-β-D葡萄糖苷和大黄素的含量,简便易行、准确、重复性好,可为维血宁的质量控制研究提供一种可靠的参考方法。Objective: To establish a high performance liquid chromatographic method for simultaneous determination of the contents of Polydatin, Emodin-8-O-β-D-glycosidase and Emodin in Weixuening. Methods: HPLC was applied with the chro- matographic condition as follows; the chromatographic column was Kromasil C18 Column (4.6 mm×250 mm, 5 μm); acetonitrile-0.1% H3PO4 as the mobile phrase with gradient elutian; flow rate was 1.0 mL.min-1; the detection wave length was 288 nm; temperature was 30 ℃. Results: Linearity relationships were 0.09-0.72 μg (r=0.9984), 0.4825-3.86 μg (r=1.0000) and 0.2675- 2.14 μg (r=1.0000), respectively of Polydatin, Emodin-8-O-β-D-glycosidase and Emodin, as well as the average recoveries of 98.9% (RSD%=2.8%),99.6% (RSD%=0.16%) and 98.1% (RSD%=0.44%). Conclusion: The results showed that the method is simple, accurate and repeatable and that it is suitable for simultaneous determining the contents of Polydatin, Emodin-8-O-β- D-glycosidase and Emodin in Weixuening.
