摘 要:利用TAS-ELISA方法对采自云南不同烟草种植区的710份花叶病样进行检测,其中546份样品与TMV抗体呈阳性反应。根据地理分布的差异,选择了38个代表性分离物进行TMV外壳蛋白基因的IC-RT-PCR扩增、克隆和测序。测序分析表明,38个TMV cp基因核苷酸序列相似性在95%~100%之间,其推导的氨基酸序列同源性为97.5%~100%。云南不同TMV分离物与TMV-U1、TMV-Fujian核苷酸相似性大于90%,其系统关系与TMV-U1和TMV-Fujian分离物聚为一簇,研究表明云南38个烟草分离物均属于TMV-U1株系。710 samples of tobacco plants showing mosaic symptom were collected from different tobacco-growing areas in Yunnan province. 546 of the 710 samples were detected TMV antigen positive by TAS-ELISA. According to geographical distribution, 38 isolates were amplified, cloned and sequenced by IC-RT-PCR for TMV coat protein gene. Result showed that nucleotide sequences for coat protein of these 38 isolates were 95% to 100% similar while amino acid sequences were similar 97.5%-100%. The similarity among different TMV isolates in Yunnan and TMV-U1, TMV-Fujian were relatively high in nucleotide sequences of coat protein gene. The phylogenetic tree of TMV isolates showed TMV isolates in Yunnan share certain evolutionary relationship with TMV-U1 strain.
