摘 要:大口黑鲈(Micropterus salmoides)“优鲈1号”是经多年选育获得的生长性状改良的养殖新品种,与非选育大口黑鲈相比,生长速度提高了17.8%一25.3%。为了研究具有生长相关优势基因型经聚合后对生长性状的改良起的贡献作用大小,本研究从大口黑鲈“优鲈1号”与非选育大口黑鲈群体中各随机挑选140ind,应用STR基因分型技术检测8个生长相关分子标记在这两个群体中的分布情况。8个生长标记中有4个微卫星位点,分别为JZL60、JZL67、MisaTpw76和MisaTpwll7,其余4个是单核苷酸多态性(single nucleotide polymorphism,SNP)位点,分别位于胰岛素样生长因子-I(insulin—like growth factor—I,IGF-I)、肌肉生长抑制素(myostatin,MSTN)、垂体特异性转录因子(pituitary specific transcription factorl,POUlFl)和生长抑素前体(preprosomatostatin,PSSIII)基因上。结果表明,“优鲈1号”与非选育群体中优势基因型的数量为1—6个,两个群体含有的平均数量分别为2.606、2.312,表明人工选育在一定程度上聚合了优势基因型,研究结果为下一步生长性状相关标记的聚合及辅助选育应用提供参考依据。The "Youlu No. 1" largemouth bass (Micropterus salmoides) is an excellent variety that has been improved in growth through selective breeding. The growth rate increased by 17.8% -25.3% compared with non-breeding largemouth bass. To investigate and analyze the contribution of growth-related advantage genotypes pyramiding, the research randomly selected 140 largemouth bass from "Youlu No. 1 " breeding population and normally cultured population respectively. Using the technology of STR genotyping, 8 growth- related molecular markers were detected in the two populations. For the 8 molecular markers, there were 4 sites of microsatellites, including JZL60, JZL67, MisaTpw76 and MisaTpw117, and 4 sites of single nucleotide polymorphism (SNP) that located in IGF-I, MSTN, POU1F1 and PSSIII gene. The results showed that each individual in the "Youlu No. 1 " population and non-breeding population had 1 to 6 advantage genotypes, with the average of 2. 606 and 2. 312 respectively. It is suggested that the artificial selection can enrich the advantage genotypes to some extent. Our results will provide useful information for further gene pyramiding breeding using growth-related markers.
