调肝益气通络方药物血清对骨髓间充质干细胞体外增殖的影响

摘　　要：目的：通过观察调肝益气通络方对大鼠间充质干细胞增殖能力的影响，从而评价该方的促增殖能力。方法：腹腔注射“CCl4-橄榄油”混悬液制备Wistar小写大鼠（35只）肝纤维化模型；其中30只灌胃给予调肝益气通络方（低、中、高3个剂量）1个月，制备含药血清；另外5只用于MSC细胞的分离与培养；用含药血清诱导MSC向肝细胞的分化连续7d；在各检测点进行形态学观察及通过MTT对细胞增殖情况进行评价。结果：各浓度含药血清在加入1—7d后，其细胞数量均比常规培养组增多，并同用药剂量呈正比，浓度越大细胞数量越多，细胞数随着时间的推移而不断增大。结论：调肝益气通络方对大鼠骨髓里的间充质肝细胞的增殖能力具有显著的促进作用。Objective：By observing the effect of Tiaogan Yiqi Tongluo Formula on mesenchymal stem cell proliferation to evaluate the proliferation ability of the party. Methods ： Intraperitoneal injection of CCl4 - olive oil was applied for the suspension prep- aration of 35 Wistar rats models of hepatic fibrosis ;30 were given Tiaogan Yiqi Tongluo Formula （low, medium and high dose） a month to prepare containing serum; the other 5 were for isolation and culture of MSC cells ; MSC drug - containing serum-induced liver cell differentiation was for 7 days ; morphology was observed in the detection and cell proliferation was evaluated by MTr. Re- sults ： The concentration of drug - containing serum added 1 - 7 days, the number of cells was increased more than that of the con- ventional culture group, and proportional to the same dose, the greater the concentration, the more the number of cells, the number of cells increased as the time passing. Conclusion： Tiaogan Yiqi Tongluo Formula has a significant role in promoting the prolifera- tion of rat bone marrow mesenchymal quality hepatocytes.
