摘 要:目的探讨当归红芪超滤物是否对TM40D人乳腺癌荷瘤小鼠具有放射增敏效应,并探讨其效应的可能机制。方法 40只TM40D人乳腺癌荷瘤小鼠随机分为对照组、药物组、放射组和联合组,每组10只。各组小鼠待肿瘤体积为120mm3时,对照组给予0.9%NaCl溶液0.3ml/d灌胃;药物组给予当归红芪超滤物1.44g/kg灌胃;放射组给予一次性6Gy射线照射;联合组给予当归红芪超滤物1.44g/kg灌胃和一次性6Gy射线照射。各组连续灌胃30 d后,观察小鼠移植瘤重量和抑瘤率、肿瘤生长延迟时间(TGD)、肿瘤体积生长至放射治疗前3倍所需时间(TGT3)及联合组的增敏系数(EF),检测各组小鼠肿瘤组织中p53mRNA及其蛋白的表达。结果联合组小鼠移植瘤重量显著低于对照组、药物组、放射组(P<0.05或P<0.01);联合组的抑瘤率显著高于药物组和放射组(P<0.01)。联合组小鼠TGT3、TGD较药物组和放射组明显增加(P<0.05或P<0.01),EF为1.95。联合组p53 mRNA及蛋白表达亦明显高于药物组、放射组(P<0.05)。结论当归红芪超滤物可以增强TM40D人乳腺癌荷瘤小鼠的放射敏感性,其机制可能是通过上调p53蛋白及其mRNA表达而发挥作用。Objective To study the radiosensitization effect of Angelica-Radix Hedysari uhrafiltration membrane extract in TM40D human breast tumor-bearing mice and its possible mechanism. Methods Forty TM40D human breast tumor-bearing mice were randomized into the control group, drug group, radiation group and combination group, with 10 in each. The control group was gavaged with 0. 3 ml/d of normal saline, drug group was gavaged with 1.44 g/kg of Angelica-Radix Hedysari ultrafihration membrane extract, radiation group was given a one-time 6Gy ray, and combination group was given 1.44 g/kg of Angelica-Radix Hedysari uhrafiltration membrane extract and a one- time 6Gy ray when the tumor volume in mice was 120ram3. The tumor weight, tumor-inhibition rate, tumor-growth delay (TGD), time required for tumor volume growth to 3 times as before radiation (TGT3) and enhancement factor (EF) in the combination group was observed after 30 days of administration. The expressions of p53 mRNA and its protein were detected. Results The tumor weight in the combination group was significantly lower than that in the control group, drug group and radiation group ( P 〈 0. 05 or P 〈 0. 01 ). The tumor-inhibition rate in the combination group was significantly higher than that in the drug group and radiation group (P 〈 0. 01 ). The TGT3 and TGD in the combination group was significantly increased as compared with the drug group and radiation group (P 〈 0. 05 or P 〈 0.01 ). The EF in the combination group was 1.95. The expressions of p53 mRNA and its protein in the combination group were significantly higher than those in the drug group and radiation group (P 〈 0. 05 ). Conclusion Angelica-Radix Hedysari uhrafiltration membrane extract can enhance the radiosensitivity of TM40D human breast tumor-bearing mice probably by upregulating the expression of p53 mRNA and its protein.
