摘  要:研究日粮中性洗涤纤维(NDF)水平对断奶獭兔肠粘蛋白基因MUC1与MUC2表达的影响,用以粘蛋白反映的肠黏膜免疫状况来确定最佳的日粮NDF添加水平。试验选用56只体质量相近的35日龄断奶幼兔,随机均分4组,分别饲喂含NDF 29%、33%、37%、41%的日粮,饲喂28d后,分别采集十二指肠、空肠、回肠、盲肠样品,-80℃保存,对MUC1及MUC2进行RT-PCR实时荧光定量。结果表明,十二指肠、空肠、盲肠中的MUC1各自表达受NDF水平影响不显著(P>0.05),回肠中NDF 37%组MUC1表达量较NDF 29%组表达量上调4.95倍(P<0.05),其余组无差异(P>0.05);空肠、回肠、盲肠中的MUC2各自表达受NDF水平影响不显著(P>0.05),十二指肠中NDF 33%组、NDF 37%组MUC2表达量较NDF 29%组依次上调2.65倍(P<0.05)、3.13倍(P<0.05),其余组无差异(P>0.05);十二指肠与回肠有较高的MUC1及MUC2异常表达概率,空肠MUC1及MUC2异常表达概率较低,盲肠MUC1及MUC2异常表达概率均为0.00;NDF 37%组MUC1及MUC2异常表达概率在4组中最高,分别为0.30、0.23;NDF 33%组MUC1及MUC2异常表达概率在4组中最低分别为0.10、0.08。该研究显示,MUC1与MUC2肠表达量可反映肠道健康状态,獭兔十二指肠与回肠是病变感染易发区;日粮纤维可以影响十二指肠及回肠MUC1与MUC2表达,日粮添加33%的NDF有助于塑造较好的肠道内环境。The paper studies the influence of ration (NDF) to theMUC1 andMUC2 expression level of the weaned Rex rabbit and determines the optimal addition level for its ration NDF with the intesti- nal mucosal immune status. 56 weaned young rabbits with similar body weight at 35days were selected and randomly divided into 4 groups, and they were fed rations COntaining NDF 29%, 33%, 37%, 41%. 28 days later, duodenum, jejunum, ileum, cecum samples were selected 8nd frozen under --80 ℃ condition. Real-time fluorescence quantitative RT-PCR were conducted for defection the ex pression ofMUC1 andMUC2 . The results showed that: NDF level had no significant effect on the expression of MUC1 in duodenum, jejunum and cecum (P〈0.05); Compared to NDF 29% group NDF 37% group of ileum raised 4.95 times (P〈0.05), and the other group had no difference (P〈 0.05) ; NDF had no significant effect on the expression level of MUC2 in Jejunum, ileum and cecum(P〉0.05), MUC2 expression value of NDF 33% and NDF 37% group of duodenum raised 2. 65 times (P〈0.05), 3.13 times (P〈0.05), and the other group had no difference (P〉0.05) ; Duode nal and ileum had higher MUC1 and MUC2 abnormal expression probability, while the probability of jeiunumMUC1 and MUC2 was lower, and the probability of appendix MUC1 and MUC2 was 0.00% MUC7 andMUC2 abnormal expression probability of NDF 37%group were 0. 30 and 0. 23, which were the highest in the 4 groups; While the probability ofMUC1 andMUC2 in NDF 33% group were 0.10 and 0.08, which were the lowest in the 4 groups. The study showed that intestinal expression of MUCI and MUC2 could reflect the intestinal health, and Rex Rabbit duodenum and ileum were prone to infection; Ration fiber could affect the expression of MUC1 , MUC2 inside duodenum and ileum. 33 % NDF ration was helpful to shape a good intestinal environment.
