摘 要:为优化胚性愈伤的培养条件,建立甘薯高效胚性悬浮培养试验操作体系,以徐薯55-2、徐薯18、徐薯22、徐薯28、徐薯29和徐紫薯5号等6个甘薯品种(系)茎尖诱导的胚性愈伤组织为材料,通过比较不同品种的胚性愈伤组织诱导率、愈伤状态等的差异,筛选出愈伤诱导率高,愈伤状态好,体胚诱导率及成苗率高的品种徐薯55-2。通过测定细胞沉降体积(SCV)研究徐薯55-2胚性细胞悬浮系的接种密度、接种量、摇床转速对胚性悬浮细胞生长的影响。结果表明,徐薯55-2胚性细胞悬浮系适宜培养条件为接种密度1∶25,接种量1.4mL,摇床转速100r/min。Embryogenic calli induced from shoot apices of 6 cultivars of sweetpotato Elpornoea batatas (L). Lam.-1, including Xushu 18, Xushu 22, Xushu 28, Xushu 29, Xushu 55-2, and Xuzishu 5 were used to optimize the culturing conditions of embryogenic callus and establish efficient experiment operating system of sweetpotato embryogenic suspension culture. Xushu 55-2 performed better with high efficiency, good callus status, high somatic embryogenesis and plant regeneration rate compared with other cultivars. Affecting factors of embryogenic suspension callus of Xushu 55-2 were studied by measuring its sedimented cell volume (SCV). The results developed the optimal culture conditions of embryogenic cell suspension. The inoculation density was 1 : 25, inoculation quantity was 1.4 mL, rotation speed was 100 r/min.
