摘  要:目的观察丹参酮ⅡA(TanshinoneⅡA,TanⅡA)对压力负荷增加大鼠心肌组织核因子-κB(NF-κB)表达的影响,并探讨其对心肌纤维化的保护作用。方法采用腹主动脉缩窄术制备压力负荷心肌纤维化模型,术后存活32只大鼠随机分成4组(n=8),假手术组、模型组、TanⅡA组(20 mg·kg-1·d-1)及阳性对照药物卡托普利组(100 mg·kg-1·d-1)。术后4周成功造模并开始给药,疗程为4周。8周后检测左心室重量指数、心肌组织病理学、心肌羟脯氨酸含量以及心肌组织NF-κB p65蛋白含量。结果与假手术组比较,模型组大鼠心室治疗指数和左心室质量指数明显升高(P<0.01);与模型组比较,TanⅡA则显著降低(P<0.01)。与假手术组比较,模型组大鼠心肌羟脯氨酸含量明显升高(P<0.01);与模型组比较,TanⅡA组则显著降低(P<0.01)。与假手术组比较,模型组大鼠心肌组织NF-κB p65表达显著升高(P<0.01);与模型组比较,TanⅡA组则显著降低(P<0.01)。结论 TanⅡA能抑制心肌肥厚,减轻心肌纤维化,此作用可能与部分下调NF-κB表达有关。Objective To observe the effect of tanshinone Ⅱ A (Tan ⅡA) on expression of myocardial NF-κB in rats with pressure overload, and to explore its protective effects on myocardial fibrosis. Methods The pressure overload myocardial fibrosis models were established by abdominal aorta constriction in Sprague-Dawley ( SD ) rats, these rats were randomly divided into four groups : sham - operation group, model group, Tan 11 A group ( 20mg ·kg- 1 . d - 1 ) and positive control group (captopril 100mg·kg-1 . d-1) ,with 8 rats in each group. 4 weeks after the operation, the animal models were successfully established, then the rats were given drugs, with a course of 4 weeks. After 8 weeks, left ventricular mass index, myocardial tissue pathological changes,myocardial hydroxyproline content,myocardial NF-κB p65 expression were detected in rats of the four groups. Results As compared with those in sham - operation group, cardiac mass index and left ventricular mass index were significantly increased in model group ( P 〈 0. 01 ) ; as compared with those in model group, these indexes were obviously decreased in Tan II A group ( P 〈 0. O1 ). As compared with that in sham - operation group, myocardial hydroxyproline content was significantly increased in model group ( P 〈 0.01 ), however, as compared with that in model group,myocardial hydroxyproline content was obviously decreased in Tan ⅡA group ( P 〈0.01 ). As compared with those in sham -operation group, NF-κB p65 protein expression levels in myocardial tissues were significantly increased in model group ( P 〈0.01) ,however,as compared with those in model group,which were significantly decreased in Tan Ⅱ A group ( P 〈0. 01 ). Conclusion Tan II A can inhibit cardiac hypertrophy, relieve myocardial fibrosis, and the effect may be related to down - regulation of the expression of NF-κB.
