摘 要:以芳香醛、乙酰乙酸乙酯(或甲酯)、碳酸铵为原料,水为溶剂,聚乙二醇-400为相转移催化剂,一锅法合成了一系列具有生物和药物活性的Hantzsch酯1,4-二氢吡啶。探讨了原料的用量、反应温度、溶剂和相转移催化剂的量对Hantzsch反应的影响,利用薄板层析(TLC)确定了反应时间,同时考察了其他铵盐对反应的影响,得到了较理想的实验条件。该方法具有产率高、污染少、环境友好等特点。产物结构经IR、^1HNMR分析确认。A series of Hantzsch esters 1,4-dihydropyridines with bioactive and biomedical property were synthesized by one-pot method in water using aromatic aldehyde, ethyl (methyl) acetoacetate and ammonium carbonate as starting materials and polyethylene glycol-400 (PEG- 400) as phase transfer catalyst. The effects of different factors were investigated such as ratio of raw materials, reaction temperature, solvent and ratio of phase transfer catalyst. Reaction time was confirmed using thin layer chromatography (TLC), ammonium salts were also studied and the optimum experiment conditions were decided. The method has the advantages of good yields, less pollution and friendly environment. The structures of final products were characterized by PT-IR and ^1H NMR.
